结直肠癌中SOX4蛋白表达与肿瘤出芽关系及临床意义 被引量:2

Correlation between SOX4 protein and tumor budding in patients with colorectal carcinoma and its clinical significance

摘要 目的在结直肠癌(CRC)中检测SOX4蛋白表达和肿瘤出芽(TB)的分级情况,并分析其与CRC临床病理因素及预后的关系。方法采用免疫组织化学方法(Immunohistochemical,IHC)检测126例CRC组织标本SOX4蛋白和E-cadherin蛋白表达情况,观察TB的分级情况,并采用卡方检验、Spearman及Cox回归等统计学方法分析其临床病理因素及预后的关系。结果在126例CRC组织中,SOX4蛋白阳性表达率为54%(68/126),肿瘤出芽率为40.5%(51/126);SOX4蛋白与肿瘤淋巴结转移相关(P<0.05),TB分级与肿瘤分化程度、浸润深度、淋巴结转移和远处转移相关(P<0.05);Spearman相关性分析发现SOX4蛋白表达与TB呈正相关(r=0.228,P=0.01),多因素生存分析发现SOX4和TB是CRC独立预后因子,SOX4蛋白阳性表达和TB的患者5年总体生存率显著降低(P<0.05)。结论SOX4蛋白和肿瘤出芽与CRC侵袭和转移密切相关,三者联合检测可以作为判断CRC转移和预后的标志物。 Objective This study aimed to detect the expression of SOX4 protein and tumor budding(TB)stage in colorectal cancer(CRC),and analyze their relationship with clinicopathological factors and prognosis.Methods Immunohistochemical(IHC)staining was used to detect the positive expression of SOX4 protein and E-cadherin in 126 CRC tissue samples.Tumor budding stage was observed.Chi-square test,Spearman and Cox regression were used to analyze the relationship between clinicopathological factors and prognosis.Results SOX4 protein positive expression rate in CRC tissues was 54%(58/126),and the rate of tumor budding was 40.5%(51/126).The positive expression of tumor budding of SOX4 in CRC was correlated with Lymph node metastasis(P<0.05).The positive expression rate of tumor budding in CRC was related to pathological differentiation,depth of invasion,Lymph node metastasis and distant metastasis(P<0.05).Spearman correlation analysis found that SOX4 protein expression was positively correlated with tumor budding(r=0.228,P=0.01).Multivariate survival analysis revealed that the expression of SOX4,tumor budding were independent prognostic factors for patients with CRC.Patients with positive SOX4 protein expression and TB had a significantly reduced 5-year overall survival(P<0.05).Conclusions The expression of SOX4 and tumor budding is closely related to tumor metastasis and invasion in CRC.Combined detection of these factors may used as a marker that predicting the prognosis and metastasis in CRC patients.
