HPLC-DAD法同时测定滋阴清热降糖丸中9种成分

Simultaneous determination of nine constituents in Ziyin Qingre Jiangtang Pills by HPLC-DAD

目的建立HPLC-DAD法同时测定滋阴清热降糖丸(北沙参、麦冬、地黄)中芒果苷、毛蕊花糖苷、鲁斯可皂苷元、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚、苍术素的含量。方法该药物甲醇提取液的分析采用InertSustain C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相乙腈-甲醇-0.1%磷酸,梯度洗脱;体积流量1.0 mL/min;柱温30℃;检测波长254、280、340 nm。结果9种成分在各自范围内线性关系良好(r≥0.9995),平均加样回收率95.86%~100.91%,RSD 1.65%~2.80%。结论该方法简便可靠,重复性好,可用于滋阴清热降糖丸的质量控制。

AIM To establish an HPLC-DAD method for the simultaneous content determination of mangiferin,verbascoside,ruscosapogenin,aloe-emodin,rhein,emodin,chrysophanol,emodin methyl ether and atractylodes in Ziyin Qingre Jiangtang Pills(Glehniae Radix,Ophiopogonis Radix,Rehmanniae Radix,etc.).METHODS The analysis of methanol extract of this drug was performed on a 30℃thermostatic InertSustain C_(18) column(250 mm×4.6 mm,5μm),with the mobile phase comprising of acetonitrile-methanol-0.1%phosphoric acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelengths were set at 254,280,340 nm.RESULTS Nine constituents showed good linear relationships within their own ranges(r≥0.9995),whose average recoveries were 95.86%-100.91%with the RSDs of 1.65%-2.80%.CONCLUSION This simple,reliable and reproducible method can be used for the quality control of Ziyin Qingre Jiangtang Pills.
