基于网络药理学方法探索新加交泰丸治疗失眠的机理

Exploring the mechanism of Xinjiajiaotai pill in the treatment of insomnia based on network pharmacology method

目的采用网络药理学方法探索新加交泰丸治疗失眠的机理。方法在中药系统药理学数据库与分析平台(TCMSP)、中国天然产物化学成分库等数据库检索新加交泰丸相关中药化学成分及潜在靶点。通过GeneCards、OMIM数据库挖掘失眠相关作用靶点,两者取交集筛选,再进行富集分析。结果筛选出新加交泰丸有效生物活性成分27个,预测出中药活性成分相关靶点235个,失眠相关靶点2323个。通过韦恩图取交集得到新加交泰丸在失眠障碍治疗中作用靶点110个。KEGG富集分析共得到97条通路,主要涉及相关癌症通路、TNF信号通路、toll样受体信号通路、NF-κB信号通路、cGMP-PKG信号通路、PI3K-Akt信号通路以及神经系统相关通路,如胆碱能突触通路、多巴胺能神经通路等。结论新加交泰丸可以多通路、多靶点交互作用,通过调节炎症因子、抗肿瘤、调节神经内分泌等相关通路,发挥改善睡眠的作用。

Objective To explore the mechanism of Xinjiajiaotai pill in the treatment of insomnia by network pharmacology method.Methods The chemical components and potential targets of Xinjiajiaotai pill were searched in traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)and Chinese natural product chemical composition database,etc.The targets related to insomnia were mined through GeneCards and OMIM databases,the intersection of the two was screened,and then enrichment analysis was carried out.Results Twenty-seven effective bioactive components of Xinjiajiaotai pill were screened,235 related targets of active components of traditional Chinese Medicine and 2323 insomnia related targets were predicted.Through the intersection of Venn diagram,110 targets of Xinjiajiaotai pill in the treatment of insomnia were obtained.A total of 97 pathways were obtained by KEGG enrichment analysis,mainly involving related cancer pathway,TNF signal pathway,toll like receptor signal pathway,NF-κB signal pathway,cGMP-PKG signal pathway,PI3K-Akt signal pathway and nervous system related pathways,such as cholinergic synaptic pathway,dopaminergic nerve pathway,etc.Conclusion Xinjiajiaotai pill can interact with multiple pathways and multiple targets,and improve sleep by regulating inflammatory factors,anti-tumor,neuroendocrine and other related pathways.
