摘要
目的:建立一种分离纯化达乌里芯芭中毛蕊花糖苷和异毛蕊花糖苷的方法。方法:以70%乙醇加热回流提取达乌里芯芭全草,依次采用石油醚、乙酸乙酯、正丁醇萃取后,经高效液相色谱法确认毛蕊花糖苷和异毛蕊花糖苷主要存在于正丁醇部位。采用离心分配色谱法,以乙酸乙酯∶正丁醇∶水(4∶1∶5)为溶剂系统,上相为固定相,下相为流动相,流速为3 mL/min,转速为1500 r/min,降相模式运行,对正丁醇部位中的毛蕊花糖苷和异毛蕊花糖苷进行分离。结论:离心分配色谱法可以高效、快速地分离纯化达乌里芯芭中的毛蕊花糖苷和异毛蕊花糖苷。
Objective:To establish a method for isolation and purification of acteoside and isoacteoside from Cymbaria dahurica L.Methods:The whole plant of Cymbaria dahurica L.was extracted by heating and refluxing with 70%ethanol.After extraction with petroleum ether,ethyl acetate,and n-butanol,high performance liquid chromatography(HPLC)was used to confirm that acteoside and isoacteoside mainly present in n-butanol.Then,centrifugal partition chromatography was adopted,ethyl acetate:n-butanol:water(4∶1∶5)was the solvent system,the upper phase was the stationary phase,the lower phase was the mobile phase,the flow rate was 3 mL/min,the rotation speed was 1500 r/min,and the operation was in the descending phase of centrifugal partition chromatography for separating acteoside and isoacteoside.Results:Centrifugal partition chromatography can efficiently and quickly isolate and purify the acteoside and isoacteoside from Cymbaria dahurica L.
