摘要
目的观察补肾活血方干预后宫腔粘连(IUAs)大鼠子宫内膜组织形态变化,探讨补肾活血方对IUAs大鼠子宫内膜损伤修复的机制。方法选取清洁级性成熟未孕雌性8~10周龄SD大鼠70只,适应性饲养后,按随机数字表法分为4组:空白对照组和模型对照组每组20只,补佳乐组和补肾活血组每组15只。采用机械损伤加感染双重损伤造模法建立IUAs模型大鼠,补肾活血组和补佳乐组分别根据大鼠体重给予相应剂量的药物灌胃。灌胃处理后第10天,每组处死10只大鼠,取材进行苏木素-伊红(HE)染色检查,观察内膜形态改变,比较各组大鼠内膜厚度及腺体数量,在扫描电子显微镜下观察子宫内膜结构和胞饮突形态;每组剩余5只雌性大鼠进行合笼,妊娠第5天时处死大鼠,采用伊文思蓝染色后比较各组大鼠子宫内胚胎着床数。结果HE染色观察发现,补肾活血组、补佳乐组子宫内膜双重损伤后,子宫内膜形态较模型对照组改善明显,但并未恢复至空白对照组水平。各组子宫内膜厚度及腺体数量比较,补肾活血组、补佳乐组显著高于模型对照组,且补肾活血组显著高于补佳乐组(P<0.05)。扫描电镜观察发现,补肾活血组较补佳乐组能有效改善子宫内膜绒毛结构及促进胞饮突发育,且明显优于模型对照组,但不及空白对照组。胚胎着床数比较,补肾活血组、补佳乐组显著高于模型对照组,且补肾活血组显著高于补佳乐组(P<0.05)。结论采用感染和机械刮宫双重损伤可以成功建立IUAs大鼠模型;给予补肾活血方治疗可以有效改善IUAs大鼠子宫内膜厚度,促进内膜中腺体数量恢复及胞饮突发育,有利于胚胎着床。
Objective:To observe the morphological changes of endometrium in rats with intrauterine adhesion(IUAs)after the intervention of Bushen Huoxue Recipe,and explore the mechanism of Bushen Huoxue Recipe on the repair of endometrial injury in IUAs rats.Methods:Seventy mature non-pregnant female SD rats with the clean grade aged 8-10 weeks were selected.The rats were divided into four groups after adaptive feeding:blank control group and model control group,20 rats for each group;Progynova group and Bushen Huoxue group,15 rats for each group.The IUAs model rats were established by the double injuries caused by mechanical injury and infection.The rats in Bushen Huoxue group and Progynova group were given corresponding doses of drugs by gavage according to the body weight of rats.On the 10^(th) day after gavage,10 rats in each group were killed and the materials were taken.The pathological changes of endometrium were observed by HE.The endometrial thickness and the number of glands were compared among the groups.The structure of endometrium and the morphology of pinopodes were observed under scanning electron microscope.The remaining 5 female rats in each group were caged,and the rats were killed on the fifth day of pregnancy.Scanning electron microscope was used to observe the endometrial ultrastructure and pinopodes.Evans blue staining was used to compare the number of embryo implantation sites in uterus of rats in each group.Results:HE staining found that the endometrial morphology after the double injury of endometrium was significantly improved in Bushen Huoxue group and Progynova group compared with the model control group,but it did not return to the level of blank control group.The endometrial thickness and the number of glands in Bushen Huoxue group and Progynova group were significantly higher than those of the model control group.The endometrial thickness and the number of glands in Bushen Huoxue group were significantly higher and more than that in Progynova group(P<0.05).Scanning electron microscope observation found that Bushen Huoxue Recipe effectively improved the structure of endometrial villi and promoted the development of pinopodes compared with the Progynova group,which was significantly better than the model control group,but was inferior to the blank control group.The number of embryos implantation sites in Bushen Huoxue group and Progynova group were significantly more than that in model control group,and the number of embryo implantation sites in Bushen Huoxue group was significantly more than that in Progynova group(P<0.05).Conclusions:The IUAs model rats can be successfully established by double injuries caused by mechanical injury and infection.Bushen Huoxue Recipe can improve the endometrial thickness,promote the recovery of glands and the development of pinopodes in the endometrium,which is conducive to embryo implantation in IUAs rats.
