摘要
试验旨在通过体外酪氨酸酶抑制模型方法评价经微波辅助醇提法提取后不同溶剂(石油醚、乙酸乙酯、正丁醇、水)萃取的山银花萃取物对酪氨酸酶抑制的作用。经Lineweaver-Burk双倒数法探讨抑制作用最强萃取物对酪氨酸酶活性的抑制机制。采用荧光猝灭法研究山银花萃取物对酪氨酸酶活性的猝灭方式。结果表明,山银花萃取物中石油醚相、乙酸乙酯相、正丁醇相、水相均具有一定酪氨酸酶抑制作用,其中乙酸乙酯相的抑制活性最好, IC_(50)为276.29 mg/L。
The inhibitory effect of extracts of Flos Lonicerae extracted with different solvents(petroleum ether, ethyl acetate,n-butanol and water) on tyrosinase was evaluated by in vitro tyrosinase inhibition model method. The inhibition mechanism of the most inhibitory extract on tyrosinase activity was studied by linear Weaver Burk double countdown method. The quenching mode of tyrosinase activity by Flos Lonicerae extract was studied by fluorescence quenching method. The results showed that they contain certain tyrosinase inhibition, which were the petroleum ether phase, ethyl acetate phase, n-butanol phase and water phase in the extract of Flos Lonicerae. Among them, the ethyl acetate phase had the best inhibitory activity,and the IC_(50) was 276.29 mg/L.
