摘要
以单因素试验结果为基础,对金蚕花多糖提取工艺进行响应面法优化,并采用MTT法对金蚕花多糖的体外抗肿瘤活性进行评价。结果表明,金蚕花多糖提取的最佳工艺条件为提取温度88℃、料液比1∶40、提取时间2 h,在此条件下的金蚕花多糖提取得率为22.347%,达到回归模型预测值(23.609%)的94.65%;提取条件因素对金蚕花多糖提取得率的影响按从大到小依次为料液比、提取温度、提取时间,并且提取时间分别与料液比、提取温度之间存在交互作用;金蚕花多糖对人胃癌细胞SGC-7901和人宫颈癌细胞HeLa的增殖均有很好的抑制作用,而在0.025~0.200 mg/mL浓度下对正常人胚胎肾细胞HEK293无毒害作用,表现出明显的体外抗肿瘤作用。研究结果为金蚕花的进一步研究开发提供了依据。
On the basis of single factor experiment,we optimized the extraction process of polysaccharide from Jincanhua by response surface method,and evaluated the antitumor effect of Jincanhua polysaccharide in vitro through MTT assay.The results showed that the optimum extraction material-liquid ratio was 1∶40,with an extraction temperature of 88℃and extraction time of 2 h.Under the above conditions,the extraction yield of Jincanhua polysaccharide was 22.347%,reaching to 94.65%of the predicted value in regression model(23.609%).Effects of the three factors on extraction yield of polysaccharide from Jincanhua were lined in descending order as material-liquid ratio,extraction temperature and extraction time.Moreover,the extraction time could interact with material-liquid ratio and extraction temperature respectively.Jincanhua polysaccharide inhibited the proliferation of human gastric cancer cell SGC-7901 and human cervical cancer cell HeLa significantly,and it had no toxicity on normal human embryonic kidney cell HEK293 with the tested concentration(0.025-0.200 mg/mL),which showed good antitumor effects in vitro.The results provide a basis for further research and development of Jincanhua.
