摘要
目的考察3种固定化咪唑离子液体(SILs)SiO_(2)·Im^(﹢)·Cl^(-),SiO_(2)·Im^(﹢)·BF_(4)^(-),SiO_(2)·Im^(﹢)·PF_(6)^(-)对石上柏提取液中穗花杉双黄酮(AME)的吸附和分离效果。方法用硅胶结合3种咪唑离子液体合成3种SILs;以AME的吸附率作为考察指标,筛选最佳SILs;并通过静态吸附-解析,考察吸附时间、吸附温度、固液比、石上柏提取物浓度对AME吸附率的影响,再经动态吸附-解析实验验证,筛选最佳吸附-脱附条件。结果在静态吸附与解析实验中考察了上述3种离子液体对AME吸附率的影响,得到最优SILs是SiO_(2)·Im^(﹢)·Cl^(-)。通过单因素实验,得到最佳吸附条件:吸附时间30 min、温度30℃、固液比1.0∶6 g/mL、样品溶液浓度0.2 mg/mL。动态吸附实验验证该吸附方法可行。结论SiO_(2)·Im^(﹢)·Cl^(-)对AME具有较强吸附和脱附效果,为中药活性成分的吸附与分离研究提供新的思路。
Objective To study the adsorption and separation effects of three immobilized imidazole ionic liquids(SILs)SiO_(2)·Im^(﹢)·Cl^(-),SiO_(2)·Im^(﹢)·BF_(4)^(-),SiO_(2)·Im^(﹢)·PF_(6)^(-) on the amentoflavon(AME)in the extract of Selaginella doederleinii.Methods Three kinds of SILs were preparation through the reaction of three ionic liquids with silica gel.The adsorption rate of AME was used as the key parameter to screen the best SILs.Static adsorption-analysis experiment was performed to analyze the effects of adsorption time,adsorption temperature,solid-liquid ratio,and Selaginella doederleinii extract concentration on the adsorption rate of AME.Dynamic adsorption-desorption experiment was verified and screened the best adsorption-desorption conditions.Results The effects of the three ionic liqudes on the adsorption rate of AME were investigated in the static adsorption and analysis experiments.Among these SILs,the best SILs was verified as SiO_(2)·Im^(﹢)·Cl^(-).Through single factor experiments,the best adsorption conditions were obtained:adsorption time 30 min,temperature 30℃,solid-liquid ratio 1.0∶6 g/ml,sample solution concentration 0.2 mg/ml.Dynamic adsorption experiments verified that the adsorption method was feasible.Conclusion SiO_(2)·Im^(﹢)·Cl^(-)has a strong adsorption and desorption effect on AME,which provides new ideas for the adsorption and separation of active ingredients in traditional Chinese medicine.
