摘要
目的探讨心通颗粒对柯萨奇病毒诱导的病毒性心肌炎小鼠炎症反应及心肌损伤的保护作用。方法健康雄性BALB/c小鼠150只按照随机数表分为对照组、心肌炎组、利巴韦林组、心通颗粒高、低剂量组,每组30只。利巴韦林组、心通颗粒高、低剂量组分别进行灌胃,其余两组给予生理盐水。第7天除对照组外,其余各组注射柯萨奇B组3型(CVB3)病毒,建立病毒性心肌炎小鼠模型;7~21 d,各组再次同前给药。使用酶联免疫吸附试验测定小鼠心肌肌钙蛋白I(cTnI)、肌酸激酶同工酶MB(CK-MB)、肌酸激酶(CK)、肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)、白细胞介素(IL)-4、IL-10以及心肌组织中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)含量以及CVB3 mRNA的表达量。结果与对照组相比,心肌炎组cTnT、CK、CK-MB、MDA、TNF-α、IFN-γ以及CVB3 mRNA增加(P<0.05);SOD、GSH-Px、IL-4以及IL-10降低(P<0.05)。与心肌炎组相比,心通颗粒高剂量和低剂量组cTnT、CK、CK-MB、MDA、TNF-α、IFN-γ以及CVB3 mRNA降低(P<0.05);SOD、GSH-Px、IL-4以及IL-10升高(P<0.05),且高剂量组变化更加明显(P<0.05)。结论注射CVB3病毒成功建立小鼠心肌炎模型,通过心通颗粒(尤其高剂量)干预,检测到小鼠心肌各项生化指标以及炎症反应明显减轻,可能与炎症反应被抑制相关。
OBJECTIVE To explore the effect of Xintong Granules on inflammatory response and protection of myocardial damage of the mice with viral myocarditis induced by Coxsackie virus. METHODS Totally 150 healthy male BALB/c mice were randomly divided into the control group, the myocarditis group, the ribavirin group, the high-dose Xintong granule group and the low-dose Xintong granule group, with 30 mice in each group. The ribavirin group, the high-dose Xintong granule group and the low-dose Xintong granule group were respectively treated with gavage, the other two groups were given normal saline. All of the groups were injected with Coxsackie Group B Type 3( CVB3) virus to establish viral myocarditis mouse models on Day 7 except for the control group, the groups were respectively treated with same drugs after 7-12 days. The levels of cardiac troponin I(cTnI), creatine kinase isoenzyme MB(CK-MB), creatine kinase(CK), tumor necrosis factor-α(TNF-α) and interferon-γ(IFN-γ) as well as the expression levels of malondialdehyde(MDA), superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and CVB3 mRNA in myocardial tissues were detected by using enzyme-linked immunosorbent assay. RESULTS The levels of cTnT, CK, CK-MB, MDA, TNF-α, IFN-γ and CVB3 mRNA of the myocarditis group were significantly higher than those of the control group(P<0.05);the levels of SOD, GSH-Px, IL-4 and IL-10 of the myocarditis group were significantly lower than those of the control group(P<0.05). The levels of cTnT, CK, CK-MB, MDA, TNF-α, IFN-γ and CVB3 mRNA of the high-dose Xintong granule group and the low-dose Xintong granule group were significantly lower than those of the myocarditis group(P<0.05);the levels of SOD, GSH-Px, IL-4 and IL-10 of the high-dose Xintong granule group and the low-dose Xintong granule group were significantly higher than those of the myocarditis group, and the levels of above indexes of the high-dose Xintong granule group changed more remarkably(P<0.05). CONCLUSION The myocarditis models of mice are successfully established by injecting CVB3 virus. The myocardial biochemical indexes and inflammatory response of the mice are significantly reduced through the intervention of Xintong Granules(especially the high-dose), which may be associated with the inhibition of inflammatory response.
