长链非编码RNA核富集转录体1在HBV感染相关性肝癌中的表达及意义 被引量:3

Expression of long non-coding RNA nuclear enrichment transcript 1 in liver cancer tissues of patients with HBV infection-related liver cancer and its significance

摘要 目的探讨长链非编码RNA(lncRNAs)核富集转录体1(NEAT1)在乙型肝炎病毒(HBV)感染相关性肝癌中的表达及意义。方法选择2016年2月-2019年5月河南大学淮河医院收治的97例HBV相关性肝癌患者作为研究对象,以肝癌组织及癌旁组织作为研究样本,采用实时荧光定量反转录聚合酶链式反应(qRT-PCR)检测NEAT1在组织中的表达,绘制受试者工作特征曲线(ROC)分析lncRNA-NEAT1预测预后的截点值,根据截断值将患者分为高表达组及低表达组,分析两组临床资料,进行生存分析及COX预后分析。结果 qRT-PCR结果显示,肝癌组织、癌旁组织中lncRNA-NEAT1表达量分别为3.26(2.85,5.63)、1.52(0.45,1.74),两者比较差异具有统计学意义(P<0.05);lncRNA-NEAT1预测三年无进展生存情况的曲线下面积为0.803(0.713~0.892),最佳截断值为3.16,此时敏感度、特异度分别为92.78%、60.82%;lncRNA-NEAT1表达水平与肿瘤大小、包膜侵犯、临床分期、远处转移、AFP水平等资料相关(P<0.05);NEAT1高表达患者无进展生存时间、总体生存时间低于NEAT1低表达患者(P<0.05);包膜侵犯、临床分期、远处转移、NEAT1高表达是肝癌无进展生存期的影响因素(P<0.05);包膜侵犯、远处转移、NEAT1高表达是肝癌总体生存期的影响因素(P<0.05)。结论肝癌组织存在LncRNA NEAT1表达水平的明显升高,且其水平与肝癌临床参数及预后相关。 OBJECTIVE To explore the expression of long non-coding RNAs(lncRNAs) nuclear enriched assembly transcript 1(NEAT1) in liver tissues of the patients with hepatitis B virus(HBV) infection-related liver cancer and analyze its significance. METHODS Totally 97 patients with HBV-related liver cancer who were treated in Huaihe Hospital, Henan University from Feb 2016 to May 2019 were recruited as the study subjects. The liver cancer tissues and para-cancerous tissues were collected. The expression levels of NEAT1 in the tissues were detected by using real-time fluorescent quantitative reverse transcription polymerase chain reaction(qRT-PCR). Receiver operating characteristic(ROC) curve was drawn to analyze the cut-off value of the lncRNA-NEAT1 in prediction of prognosis, the patients were divided into the high expression group and the low expression group according to the cut off value, the clinical data of the two groups were analyzed, and the survival analysis and COX analysis were conducted. RESULTS The result of qRT-PCR showed that the expression level of lncRNA-NEAT1 was 3.26(2.85,5.63) in liver cancer tissues, 1.52(0.45,1.74)in para-cancerous tissues, and there was significant difference(P<0.05). The area under curve of the lncRNA-NEAT1 was 0.803(0.713~0.892) in prediction of no progression in three years, with the optimal cutoff value 3.16, the sensitivity 92.78%, the specificity 60.82%. The expression levels of lncRNA-NEAT1 was associated with tumor size, capsule invasion, clinical stage, distant metastasis and AFP level(P<0.05). The progression-free survival time and overall survival time of the patients with high expression of NEAT1 were significantly shorter than those of the patients with low expression of NEAT1(P<0.05).The capsule invasion, distant metastasis and high expression of NEAT1 were the influencing factors for the overall survival time of the liver cancer patients(P<0.05). CONCLUSION The expression level of LncRNA NEAT1 is significantly elevated in the liver cancer tissues, and it is associated with the clinical parameters and prognosis of liver cancer.
