蛞蝓多糖的鉴定及其含量测定

Identification and determination of Limax polysaccharides

目的:测定蛞蝓多糖的单糖组成、结构和多糖含量。方法:用观察法、苏木精—伊红(HE)染色法对蛞蝓药材进行鉴别,再采用Molish、斐林反应鉴别蛞蝓多糖,薄层色谱法分析蛞蝓多糖的单糖组成,傅里叶变换红外光谱法测定蛞蝓多糖的结构,苯酚—硫酸比色法测定蛞蝓多糖的含量。结果:蛞蝓显微鉴别有特征性;Molish、斐林反应可知提取物是含有还原糖的多糖;蛞蝓多糖主要由半乳糖、葡萄糖组成,是一种含有吡喃环型糖苷键和酰胺键的酸性蛞蝓多糖,其含量为18.47%。结论:蛞蝓多糖是一种由半乳糖、葡萄糖组成的酸性多糖,多糖含量以葡萄糖计为18.47%。

Objective:To determine monosaccharide composition,structure,and polysaccharide content of Limax polysaccharides.Methods:Firstly,the observation method and HE staining method were used to identify Limax medicinal materials,then Molish and Fehling reactions were used to identify Limax polysaccharides.The thin layer chromatography(TLC) was used to analyze the monosaccharide composition of Limax polysaccharides.Fourier transform infrared spectrometry was used to determine the structure of a polysaccharide from the Limax,and phenol-sulfuric acid colorimetry was used to determine the content of the polysaccharide.Results:The microscopic identification of Limax was characteristic;Molish and Fehling reactions showed that the extract was a kind of polysaccharide containing reducing sugars;Limax polysaccharide was mainly composed of galactose and glucose,and was an acidic Limax polysaccharide with pyranose-type glycosidic bond and amide bond,with the content of 18.47%.Conclusion:Limax polysaccharide is an acidic polysaccharide composed of galactose and glucose,with a polysaccharide content of 18.47% based on glucose.
