Effects of LncRNA HOXA-AS2 on the proliferation and epithelial-mesenchymal transition of retinoblastoma cells

目的探讨长链非编码RNA HOXA-AS2(lncRNA HOXA-AS2)对视网膜母细胞瘤细胞增殖和上皮间质转化(EMT)的影响。方法选取对数期NCI-H187人视网膜母细胞瘤细胞,采用随机数表法分为lncRNA HOXA-AS2组、对照组和空白组,分别转染pc DNA3.0-lncRNA HOXA-AS2、pc DNA3.0-NC和磷酸盐缓冲液。采用qRT-PCR检测lncRNA HOXA-AS2表达,CCK-8法检测细胞增殖指数,Annexin V FITC/PI法检测细胞凋亡指数,Transwell小室检测细胞侵袭指数,Western Blot检测Vimentin表达。结果转染后24h和48h,lncRNA HOXA-AS2组细胞的lncRNA HOXA-AS2表达水平、细胞增殖指数、细胞凋亡指数、细胞侵袭指数和Vimentin蛋白相对表达水平均高于空白组和对照组,差异均有统计学意义(均P<0.05)。结论过表达lncRNA HOXA-AS2能促进视网膜母细胞瘤细胞增殖、侵袭与凋亡,利于肿瘤细胞发生EMT,抑制肿瘤细胞凋亡。

Objective To investigate the effects of LncRNA HOXA-AS2 on the proliferation and epithelial-mesenchymal transition(EMT)of retinoblastoma cells.Methods NCI-H187 human retinoblastoma cells at a logarithmic growth were selected and randomly divided into three groups,say,a LncRNA HOXA-AS2 group,a control group and a blank group.These three groups were transfected with pc DNA3.0-lncRNA HOXA-AS2,pc DNA3.0-NC and phosphate buffer,respectively.Quantitative reverse transcription polymerase chain reaction(qRT-PCR)was used to detect the expression of LncRNA HOXA-AS2.Cell counting kit-8(CCK-8)was used to detect cell proliferation index.Annexin V-FITC/PI was used to detect cell apoptosis index.Transwell chambers were used to detect cell invasion index.Western blot was used to detect Vimentin expression.Results At 24 h and 48 h after the transfection,the expression level of LncRNA HOXA-AS2,cell proliferation index,cell invasion index,and relative expression level of Vimentin protein were higher in the LncRNA HOXA-AS2 group than those in the blank group and the control group(all P<0.05),and the apoptosis indexes were lower than in the blank group and the control group(all P<0.05).Conclusion Over-expression of LncRNA HOXA-AS2 can promote the proliferation and invasion of retinoblastoma cells,which is beneficial to the occurrence of EMT of tumor cells and inhibits tumor apoptosis.
