摘要
笔者通过测定对羟基自由基(·OH)和超氧阴离子(O_(2)^(-)·)的清除率评价异叶青兰黄酮与多糖混合物的体外协同抗氧化作用。采用ELI SA法测定灌服异叶青兰黄酮与多糖及其混合物的小鼠血清中超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)含量,评价异叶青兰黄酮与多糖混合物的体内协同抗氧化作用。结果表明,异叶青兰黄酮与多糖及其混合物对·OH和O_(2)^(-)·均具有一定的清除作用,异叶青兰黄酮与多糖的混合物的清除效果优于异叶青兰黄酮与多糖;不同剂量组异叶青兰黄酮与多糖及其混合物均能提高小鼠血清中SOD和GSH-Px含量,降低MDA含量,其中200 mg/kg异叶青兰多糖+200 mg/kg异叶青兰黄酮提高SOD和GSH-Px含量,降低MDA含量能力最强。说明异叶青兰黄酮与多糖之间存在明显的协同抗氧化效应,且呈剂量依赖性。
[Objective]The aim of this experiment was to study the synergistic antioxidant effect of flavonoids and polysaccharides from Dracocephalum heterophyllum Benth.[Methods]The synergistic antioxidant activity of flavonoids and polysaccharide mixture of Dracocephalum heterophyllum Benth in vitro was evaluated by determining the scavenging rates of the hydroxyl radir-al(·OH)and superoxide anion(O_(2)^(-)·);The contents of superoxide dismutase(SOD),glutathione peroxidase(GSH-PX)and malondialdehyde(MDA)in serum of mice fed with isoflavones and polysaccharides and their mixture were determined by ELISA to evaluate the synergistic antioxidant effect of isoflavones and polysaccharides in vivo.[Results]Flavonoids and polysaccharides and their mixes had certain scavenging effects on·OH and O_(2)^(-),and the scavenging effect of the mixture of flavonoids and polysaccharides was better than that of the mixture of flavonoids and polysaccharides;The contents of SOD and GSH-Px in serum of mice in different dosage groups were increased by flavonoids,polysaccharides and their mixtures,while the contents of MDA were decreased.Among them,200 mg/kg polysaccharide's and 200 mg/kg flavonoids increased the contents of SOD and GSH-Px,and had the strongest ability to reduce the content of MDA.[Conclusion]There were significant synergistic antioxidant effects between flavonoids and polysaccharides of Dracocephalum heterophyllum Benth in a dose-dependent manner.
