摘要
为了比较鹿流行性出血病ELISA(酶联免疫吸附试验)抗体检测和琼脂凝胶免疫扩散试验(AGID)抗体检测两种方法的优缺点,以及相同方法下不同试剂盒的特点,首先将ELISA抗体检测试剂盒ID、KE及琼扩试剂盒XI、VM,按照1:2~1:512稀释的强阳性、弱阳性、阴性种牛血清进行对比检测试验。然后将所有试剂盒对已知结果的进境种牛500份血清样本进行检测试验。结果表明:琼扩试剂盒XI、ELISA抗体检测试剂盒ID分别在强阳性血清1:16、1:64稀释度下开始呈现阴性;ELISA抗体检测试剂盒ID、KE分别在强阳性血清1:32、1:128稀释度以后呈现阴性,但KE试剂盒在1:128稀释度中的检测结果不一致,ID和KE试剂盒重复性试验的RSD分别在1.19%~9.48%和0.55%~241.5%之间,即KE较ID具有更高的灵敏度,但检测的重复性较差。500份血清样本检测中,只有KE试剂盒检测出现7个假阳性。本研究结果表明,ELISA检测试剂盒较琼扩试剂盒具有更高的灵敏度且ID试剂盒具有更好的重复性,在鹿流行性出血病的诊断检测中具有良好的实用价值。
To compare the advantages and disadvantages of ELISA kit and AGID kit of Epizootic hemorrhagic disease(EHD),we tested ID and KE kits which belong to ELISA and XI and VM kits which belong to AGID with serums diluted in the range of 1:2 to 1:512. Positive, weak positive and negative serums were all included. Then 500 bovine serums which were part of retained samples were examined with known results. The results were as follows: outcomes of XI and ID kits became negative when the positive serum dilution came to 1:16, 1:64 respectively. Results of ID and KE kits turned to negative after 1:32, 1:128 of positive serum dilution respectively. However, the test results of KE kits in the 1:128 dilution were inconsistent. The RSD of ID and KE were in the range of 1.19%-9.48% and 0.55%-241.5%, whice means KE had better sensitivity than ID but poor repeatability. In the research of 500 bovine serums, KE was the only kit which had 7 false positives. Therefore, ELISA is more sensitivity than AGID and ID has better repeatability than KE in EHD testing of bovine serums.
