固元颗粒对口蹄疫疫苗免疫小鼠免疫功能的影响研究 被引量:1

摘要 为了探讨固元颗粒对口蹄疫疫苗免疫小鼠免疫功能的影响,试验将体重相近的150只Balb/c小鼠随机均分为5组,每组30只,分别为空白对照(BC)组、阳性药物对照(APS)组和固元颗粒低(GYKL1)、中(GYKL2)、高(GYKL3)剂量组,各组小鼠皮下注射口蹄疫疫苗,免疫两次,于免疫后不同时间每组随机取5只小鼠测定体重、免疫器官指数,血清白细胞介素(IL)-2、IL-6分泌水平及IgG、IgG亚类效价。结果表明:固元颗粒对小鼠体重无影响;与BC组比较,GYKL2组小鼠的脾脏指数和胸腺指数显著提高(P<0.05);各剂量组血清IL-2分泌水平极显著提高(P<0.01);GYKL1,2,3组小鼠口蹄疫疫苗特异性IgG效价均有所提升,灌胃3周后,GYKL1,2,3组血清IgG亚类效价均极显著高于BC组(P<0.01),GYKL1组的IgG1和IgG2b效价显著高于APS组(P<0.05)。说明固元颗粒可提高正常小鼠的免疫应答水平。 In order to explore the effect of Guyuan granules on the immune function of mice immunized with foot-and-mouth disease vaccine,150 Balb/c mice with similar body weights were randomly divided into 5 groups on average,each group of 30 Balb/c mice,namely the blank control(BC)group,the positive drug control(APS)group and the Guyuan granule low(GYKL1),medium(GYKL2)and high dose(GYKL3)groups.Mice in each group were injected subcutaneously with foot-and-mouth disease vaccine and immunized twice.At different times after immunization,5 mice from each group were randomly selected to determine body weight,immune organ index,and serum levels of IL-2,IL-6 secretion level,and IgG,IgG subclasses titer.The results showed that Guyuan granules had no effect on the weight of mice;compared with BC group,the group of GYKL2 could significantly increase the spleen index and thymus index of mice(P<0.05);GYKL1,2,3 group could significantly increase the level of IL-2 secretion in the blood(P<0.01);GYKL1,2,3 group could increase the titer of foot-and-mouth disease vaccine-specific IgG in mice;after 3 weeks of intragastric administration,the serum IgG subclasses of GYKL1,2,3 group were significantly higher than those of the BC group(P<0.01);the titer of IgG1 and IgG2 b in the GYKL1 group were significantly higher than the APS group(P<0.05).The results suggested that Guyuan granules could improve the immune response level of normal mice.
