黄芩素抑制143b骨肉瘤细胞增殖及其机制 被引量:2

Inhibition effect of baicalein on the proliferation of 143b osteosarcoma cells and its mechanism

摘要 目的:研究不同浓度黄芩素(0,25,50,100μmol/L)对143b骨肉瘤细胞增殖的抑制作用及其作用机制。方法:采用CCK-8实验观察不同浓度黄芩素(0,25,50,100μmol/L)处理143b骨肉瘤细胞对细胞增殖的抑制作用;流式细胞仪分析黄芩素对143b细胞周期分布和凋亡率的影响;Western Blot分析黄芩素对Bax、Bcl-2、Hes1和NICD1蛋白水平的影响。结果:黄芩素能时间和浓度依赖性地抑制143b细胞的增殖,诱导143b细胞周期阻滞于S期并促进骨肉瘤细胞凋亡(P<0.05)。黄芩素处理后,143b细胞中促凋亡蛋白Bax蛋白水平显著升高,抑凋亡蛋白Bcl-2蛋白水平显著降低(P<0.05);同时,细胞中Hes1和NICD1蛋白水平也显著降低(P<0.05)。结论:黄芩素能诱导143b细胞周期阻滞并能促进细胞凋亡发挥对143b细胞增殖的抑制作用,可能与抑制Notch信号通路活性相关。 Objective:To study the inhibitory effect of different concentrations of baicalein(0,25,50,and100 μmol/L,respecitvely) on the proliferation of 143 b osteosarcoma cells and its mechanism.Methods:CCK-8 experiment was used to observe the inhibitory effect of different concentrations of baicalein(0,25,50,and 100 μmol/L,respecitvely) on the cell proliferation of 143 b cells.Flow cytometry was used to analyze the effect of baicalein on 143 b cell cycle distribution and cell apoptosis.Western Blot was used to analyze the effect of baicalein on expression levels of Bax,Bcl-2,Hes1,and NICD1.Results:Baicalein could inhibit the proliferation of 143 b osteosarcoma cells in a time-and concentration-dependent manner,induce 143 b cell cycle arrest in S phase,and promote 143 b cell apoptosis(P<0.05).After baicalein treatment,the pro-apoptotic protein Bax protein level in 143 b cells increased significantly,and the anti-apoptotic protein Bcl-2 protein level decreased significantly(P<0.05).At the same time,the protein levels of Hes1 and NICD1 were also significantly reduced(P<0.05).Conclusion:Baicalein could induce cell cycle arrest and promote cell apoptosis,thus exhibiting the inhibitory effect on 143 b cells,which might be associated with inhibiting the activity of Notch signaling pathway.
