摘要
目的探究S100A9蛋白与甲状腺乳头状癌细胞增殖、迁移及侵袭的关系。方法采用Western blot检测S100A9在甲状腺乳头状癌细胞株与正常细胞中的表达。建立敲低S100A9模型,并采用MTS、细胞克隆形成、划痕实验、Transwell迁移实验及皮下成瘤实验检测敲低S100A9对甲状腺乳头状癌细胞增殖、迁移、侵袭能力的影响。结果 S100A9在甲状腺乳头状癌细胞中的表达较正常细胞中表达高。MTS法、克隆形成实验及裸鼠皮下肿瘤种植表明,敲低S100A9抑制了甲状腺乳头状细胞的增殖(P<0.05)。划痕实验、Transwell实验表明敲低S100A9抑制了甲状腺乳头状细胞的迁移和侵袭(P<0.05)。结论敲降S100A9能够在体外抑制甲状腺乳头状癌细胞的增殖、迁移及侵袭能力,在体内能够抑制皮下种植肿瘤的生长。
Objective To investigate the relationship between S100A9 expression and proliferation,migration and invasion of papillary thyroid carcinoma cells. Methods Western blot was used to detect the expression of S100A9 in papillary thyroid carcinoma cell lines and normal cells. Knockdown S100A9 model was established;MTS,cell clone formation,scratch test,transwell migration test and subcutaneous tumor formation test were used to detect the effect of knockdown S100A9 on the proliferation,migration and invasion of papillary thyroid cancer cells. Results S100A9 expression level was higher in papillary thyroid cancer cells than in normal cells. Knockdown S100A9 inhibited the proliferation of thyroid papillary cells(P<0.05). Knockdown S100A9 inhibited the migration and invasion of thyroid papillary cells(P<0.05).Conclusion Knockdown S100A9 could inhibit the proliferation,migration and invasion of papillary thyroid cancer cells in vitro,and inhibit the growth of subcutaneously implanted tumors in vivo.
