急性肺动脉高压大鼠模型建立与评价

Establishment and evaluation of rat model of acute pulmonary hypertension

摘要 目的:利用U46619持续静脉注射法构建急性肺动脉高压大鼠模型,并用曲前列尼尔观察和验证该模型。方法:SD雄性大鼠12只,完全随机分成模型组和模型+曲前列尼尔组,心导管测量各组右心室收缩压及心率的变化。结果:持续注射U46619后,两组右心室收缩压均较基础值显著升高(P<0.05),两组心率较基础值变化不大,差异无统计学意义(P>0.05)。急性肺动脉高压模型建立后,予曲前列尼尔持续注射17min后,右心室收缩压出现显著下降(P<0.05),而心率较前无明显变化(P>0.05)。结论:持续注射U46619法可以建立急性肺动脉高压大鼠模型,且方法简便有效,该模型可为肺动脉高压发病机制研究及相关药物研发提供有力的模型支持。 Objective:To establish a rat model of acute pulmonary hypertension via continuous intravenous injection of U46619,and to evaluate and verify the rat model with Treprostinil.Methods:Twelve male SD rats were randomly divided into the model group and model/trepronil group.Cardiac catheterization was used to measure the changes in right ventricular systolic blood pressure and heart rate in each group.Results:After continuous injection of U46619 in either group,the right ventricular systolic pressure was significantly higher than baseline(P<0.05),but the heart rate did not showed statistically significant change from(P>0.05).After the establishment of the acute pulmonary hypertension model,continuous injection of Treprostinil for 17 min led to significant reduction in the right ventricular systolic blood pressure(P<0.05)but not in the heart rate(P>0.05).Conclusion:Continuous injection of U46619 may readily and effectively establish a rat model of acute pulmonary hypertension,which may offer strong supports for studies on pathogenesis of pulmonary hypertension and development of therapeutic drugs.
