前列腺素D2在胃癌中的表达水平及其与临床病理特征的相关性

Expression of prostaglandin D2 in gastric cancer and its correlation with clinicopathological characteristics

目的:探讨前列腺素D2(PGD2)在胃癌患者组织中的表达水平及其与临床病理特征的相关性。方法:选取2019年1月至2020年10月蚌埠医学院第一附属医院收治的68例胃癌患者,取患者胃癌组织和癌旁正常组织。通过RT-PCR检测前列腺素D2合酶(L-PTGDS)和前列腺素D2受体2(PTGDR2)基因相对表达量;免疫组化检测L-PTGDS、PTGDR2的阳性表达情况,并分析其阳性表达情况与患者临床病理特征的相关性。结果:胃癌组织中L-PTGDS与PTGDR2相对表达量低于癌旁正常组织(P<0.05),阳性表达率高于癌旁正常组织(P<0.05);胃癌组织中L-PTGDS、PTGDR2蛋白表达情况与肿瘤大小、TNM分期、胃癌淋巴结转移及远处转移具有相关性(P<0.05)。结论:L-PTGDS与PTGDR2在胃癌组织中的低表达可能导致外周血PGD2表达降低;L-PTGDS与PTGDR2阳性表达与肿瘤大小、TNM分期、胃癌淋巴结转移及远处转移具有相关性,外周血中PGD2的水平有望成为胃癌诊断及治疗的生物标志物。

Objective:To investigate the expression of prostaglandin D2(PGD2)in gastric cancer patients and its correlation with clinicopathological characteristics.Methods:A total of 68 patients with gastric cancer admitted to the First Affiliated Hospital of Bengbu Medical College between January 2019 and October 2020 were enrolled.Gastric cancer tissues and adjacent normal tissues from the patients were collected.The relative expression of prostaglandin D2 synthase(L-PTGDS)and prostaglandin D2 receptor 2(PTGDR2)was examined by RT-PCR.The positive expression of L-PTGDS and PTGDR2 was detected by immunohistochemistry.The correlation between the positive expression and the clinical pathological characteristics of the patients was analyzed.Results:The relative expression of L-PTGDS and PTGDR2 in gastric cancer tissues was significantly lower than that in normal tissues adjacent to cancer(P<0.05).The positive expression rates of L-PTGDS and PTGDR2 in gastric cancer tissues were significantly higher than those in normal tissues adjacent to cancer(P<0.05).The expression of L-PTGDS and PTGDR2 was correlated with tumor size,TNM stage,lymph node involvement of gastric cancer and distant metastasis(P<0.05).Conclusion:The low expression of L-PTGDS and PTGDR2 in gastric cancer tissue may lead to decreased PGD2 expression in peripheral blood.The positive expression of L-PTGDS and PTGDR2 is related to tumor size,TNM stage,lymph node involvement and distant metastasis of gastric cancer.Peripheral blood level of PGD2 is promising as biomarker for the diagnosis and treatment of gastric cancer.
