摘要
目的探讨Senp2基因与c-Myc基因在不同等级胶质瘤中的表达情况,并进一步探讨Senp2基因对于胶质瘤侵袭性的影响。方法运用real-time PCR、蛋白质免疫印迹法(Western Blotting)等检测不同等级胶质瘤患者病理组织及正常脑组织中Senp2、c-Myc的表达;运用细胞培养和转染技术沉默U87恶性胶质瘤细胞系中Senp2基因的表达,观察细胞中c-Myc的表达情况。通过Transwell侵袭实验、流式细胞术、CCK8法等观察Senp2基因沉默后胶质瘤细胞侵袭性变化情况。结果胶质瘤级别越高的患者c-Myc基因表达水平越高,Senp2基因表达水平越低(P<0.05)。沉默Senp2基因后可见c-Myc基因表达水平明显升高(P<0.05)。通过Transwell侵袭实验,发现沉默Senp2基因的胶质瘤细胞侵袭性强于对照组。同时当沉默Senp2基因后,细胞处于S期的比例明显增加,CCK8法检测发现沉默Senp2基因后胶质瘤细胞的增殖能力明显增强(P<0.05)。结论 Senp2基因与c-Myc基因在不同等级胶质瘤中表达存在差异,胶质瘤等级越高的患者Senp2基因表达水平越低,而c-Myc基因表达水平越高。Senp2基因可影响胶质瘤细胞中c-Myc基因的表达,从而影响胶质瘤细胞的侵袭性。
Objective To investigate the expression of Senp2 gene and c-Myc gene in different grades of glioma,then further explore the influence of Senp2 gene on the invasiveness of glioma.Methods Real-time PCR and Western blotting were used to detect the expression of Senp2 and c-Myc in pathological tissues and normal brain tissues of patients with different grades of glioma;Senp2 gene expression was downregulated in U87 malignant glioma cell lines using cell culture and transfection technology,the expression of c-Myc in cells was observed.The invasive changes of glioma cells after downregulation of the Senp2 gene were observed by Transwell invasion experiment,flow cytometry,CCK8 method,etc.Results The c-Myc gene expression level was higher in patients with higher-grade glioma,while the Senp2 gene expression level was lower(P<0.05).After downregulation of the Senp2 gene,the expression level of c-Myc gene was significantly increased(P<0.05).Through the Transwell migration assay,it was found that the glioma cells downregulating the Senp2 gene were more aggressive than the control group.At the same time,when the Senp2 gene was downregulated,the proportion of cells in the S phase increased significantly,and the detection of CCK8 method found that the proliferation ability of glioma cells was significantly enhanced after the Senp2 gene was downregulated(P<0.05).Conclusion The expression of Senp2 gene and c-Myc gene is different in different grades of glioma.The c-Myc gene expression level is higher in patients with higher-grade glioma,while the Senp2 gene expression level is lower.Senp2 gene can affect the expression of c-Myc gene in glioma cells,thereby affecting the invasiveness of glioma cells.
