摘要
目的:探讨醋制前后南五味子抗四氯化碳(CCl_(4))致急性肝损伤大鼠的功效差异。方法:将SD大鼠随机分空白对照组、模型组、阳性对照组、南五味子高、中、低剂量组和醋制南五味子高、中、低剂量组。各给药组预防给药10天,于第10天给药结束后腹腔注射1 mL/kgCCl_(4),建立急性肝损伤模型。测定大鼠血清中谷丙转氨酶(Alanine aminotransferase,ALT)和谷草转氨酶(Aspartate aminotransferase,AST)及肝匀浆中丙二酸(Malondialdehyde,MDA)、谷胱甘肽(Glutathione,GSH-Px)、超氧化物歧化酶(Superoxide dismutase,SOD)含量,免疫组化法观察肝细胞中缺氧诱导因子-1α(HIF-1α)和核转录因子-κB(NF-κB p65)表达水平,苏木精-伊红(HE)染色观察肝脏病理组织学变化。结果:南五味子醋制前后各剂量组均能降低CCl_(4)致大鼠急性肝损伤血清ALT、AST的升高,降低肝匀浆中MDA含量,增强GSH-Px、SOD的活性,抑制肝细胞中NF-kBP65和HIF-1α表达,减轻CCl_(4)对肝组织的病理损伤。与南五味子生品比较,醋制南五味子对ALT、AST、GSH-Px、SOD影响更为显著,抑制肝细胞中NF-κB p65和HIF-1α表达和减轻CCl_(4)对肝组织的病理损伤效果明显。结论:醋制增强了南五味子对CCl_(4)致急性肝损伤保护作用。
Objective:To investigate the effect of Schisandra sphenanthera decoction pieces before and af⁃ter vinegar preparation on the effects of carbon tetrachloride(CCl_(4))-induced acute liver injury in rats.Methods:Ninety rats were randomly divided into 9 groups,which were blank control group,model group,positive control group,and high,medium and low dose groups of decoction pieces before and af⁃ter the preparation of vinegar.Prophylactic administration of each administration group for 10 days,and intraperitoneal injection of 1 mL/kg CCl_(4) stock solution after the end of administration on the 10th day to establish an acute liver injury model.Determination of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in rat serum and malondialdehyde(MDA),glutathione(GSH-Px)and superox⁃ide in liver homogenate superoxide dismutase(SOD)content,immunohistochemical method to observe the expression levels of hypoxia-inducible factor-1α(HIF-1α)and nuclear transcription factorκB(NF-κB p65)in hepatocytes,hematoxylin-I Red(HE)staining was used to observe the histopathologi⁃cal changes of the liver.Results:All the dose groups of the decoction pieces before and after the prepara⁃tion of vinegar of S.sphenanthera can reduce the increase in serum ALT and AST of CCl_(4)-induced acute liver injury in rats,reduce the content of MDA in liver homogenate,enhance the activity of GSH-Px and SOD,and inhibit the expression of hepatocytes NF-kB p65 and HIF-1α,reduce the pathological dam⁃age of liver tissue caused by CCl_(4).Compared with raw products of S.sphenanthera,vinegar-made S.sphenanthera has more significant effects on ALT,AST,GSH-Px,SOD,inhibits the expression of NFκB p65 and HIF-1αin liver cells and reduces the pathological damage of CCl_(4) to liver tissue.Conclusion:The vinegar preparation enhances the protective effect of S.sphenanthera on CCl_(4)-induced acute liver in⁃jury.
