Effects of the AgrB signal molecule on biofilm formation by Clostridium perfringens type B

摘要 目的探讨QS系统AgrB分子对B型产气荚膜梭菌生物被膜(biofilm, BF)形成过程中htrA、rpoA基因表达水平的影响。方法体外培养B型产气荚膜梭菌12、24、36、48、56及64 h时期的BF,提取其总RNA,通过实时Real-Time PCR检测不同时间段B型产气荚膜梭菌BF agrB基因对BF形成相关基因htrA、rpoA表达的调节作用。结果 B型产气荚膜梭菌在24、36、48、56及64 h 5个时间段中,QS系统agrB基因和BF rpoA、htrA基因相对表达量均呈现出先增高后降低的趋势,且在36 h达到峰值。在峰值前期,agrB、rpoA、htrA基因的表达量较高,48 h后表达量迅速下降,提示B型产气荚膜梭菌BF形成相关基因rpoA、htrA的表达受到QS系统agrB基因表达的影响。结论 AgrB信号分子对rpoA、htrA基因调控表达趋势在产气荚膜梭菌BF形成过程呈现关联性。 Objective To investigate the effect of the AgrB molecule of the QS system on the level of expression of the htrA and rpoA genes during the formation of biofilm(BF) by Clostridium perfringens type B. Methods The BF of C. perfringens type B was cultured in vitro for 12, 24, 36, 48, 56, and 64 hours, and its total RNA was extracted. The regulatory action of expression of the BF formation-related genes htrA and rpoA on the agrB gene, which is involved in BF by C. perfringens type B, was detected using real-time PCR. Results The relative expression of the agrB gene, the rpoA BF gene, and the htrA gene in the QS system initially increased and then decreased at 24, 36, 48, 56, and 64 hours, and it peaked at 36 hours. The level of expression of the agrB, rpoA, and htrA genes was higher during the early peak and decreased rapidly after 48 hours. This suggests that expression of the rpoA and htrA genes in relation to BF formation by C. perfringens type B is influenced by expression of the agrB gene in the QS system. Conclusion Trends in rpoA and htrA expression regulated by the AgrB signal molecule are related to BF formation by C. perfringens.
