Wnt/β-catenin信号通路在胎儿生长受限新生儿外周血中表达及临床意义

Expression of Wnt/β-catenin Signaling Pathway in Peripheral Blood of Neonates with Fetal Growth Restriction and its Clinical Significance

摘要 目的探究Wnt/β-连环蛋白(β-catenin)信号通路在胎儿生长受限(fetal growth restriction,FGR)新生儿外周血中表达及临床意义。方法选取分娩的73例孕妇作为研究对象,分娩后根据胎龄及出生体质量分为FGR组22例、适于胎龄儿组(AGA组)34例及大于胎龄儿组(LGA组)17例。比较各组新生儿生长参数、Wnt/β-catenin信号通路相关蛋白表达情况,并对Wnt/β-catenin信号通路相关蛋白与新生儿生长参数进行相关性分析。结果3组新生儿体质量指数(BMI)、头围、身长、胎盘质量、体质量、Wnt信号蛋白中7a(Wnt7a)、Wnt信号蛋白中7b(Wnt7b)、β-catenin、轴抑制蛋白2(Axin2)、糖原合酶激酶3(GSK-3β)比较,差异有统计学意义(P<0.05);与LGA组比较,FGR组和AGA组新生儿BMI、头围、身长、胎盘质量、体质量、Wnt7a、Wnt7b、β-catenin、Axin2、GSK-3β降低,差异有统计学意义(P<0.05);与AGA组比较,FGR组新生儿BMI、头围、身长、胎盘质量、体质量、Wnt7a、Wnt7b、β-catenin、Axin2、GSK-3β降低,差异有统计学意义(P<0.05)。FGR新生儿脐血中Wnt7a、Axin2及GSK-3β水平与新生儿生长参数呈正相关(P<0.05),而Wnt7b与新生儿身长无明显相关(P>0.05),β-catenin与头围及体质量无明显相关(P>0.05)。结论与正常新生儿比较,FGR新生儿生长发育受限,可能与Wnt/β-catenin信号通路活性降低有关。 Objective To explore the expression of Wnt/β-catenin signaling pathway in peripheral blood of neonates with fetal growth restriction and its clinical significance.Methods Seventy-three pregnant women were selected as the research subjects.After delivery,they were divided into fetal growth restriction group(FGR group,22 cases),suitable for gestational age group(AGA group,34 cases)and older than gestational age group(LGA group,17 cases)according to gestational age and birth weight.The newborn growth parameters and Wnt/β-catenin signaling pathway related protein expression were compared in each group,and the correlation between Wnt/β-catenin signaling pathway related protein and newborn growth parameters was analyzed.Results There were significant differences in BMI,head circumference,height,placental weight,body weight,Wnt7a,Wnt7b,β-Catenin,Axin2 and GSK-3βamong the three groups(P<0.05);compared with LGA group,the BMI,head circumference,body length,placental mass,body weight,Wnt7a,Wnt7b,β-catenin,Axin2 and GSK-3βof neonatal in FGR group and AGA group were decreased,the differences were statistically significant(P<0.05).Compared with AGA group,the BMI,head circumference,body length,placental mass,body weight,Wnt7a,Wnt7b,β-catenin,Axin2 and GSK-3βof neonatal in FGR group were decreased,the differences were statistically significant(P<0.05).The levels of Wnt7a,Axin2 and GSK-3βin umbilical blood of neonates with FGR were positively correlated with neonatal growth parameters,the differences were statistically significant(P<0.05),but there were no significant correlation between Wnt7b and neonatal body length(P>0.05),and there were no significant correlation betweenβ-catenin and head circumference and body weight(P>0.05).Conclusion Compared with normal neonates,the growth and development limitations of FGR neonates may be related to the decreased activity of Wnt/β-catenin signaling pathway.
