妊娠早期七氟醚全麻对仔鼠海马、顶叶皮质中干扰素诱导蛋白表达及炎性反应的影响

摘要 目的 探讨妊娠早期七氟醚全麻对仔鼠海马、顶叶皮质中干扰素诱导蛋白(AIM2)表达及炎性反应的影响。方法 将24只胎龄为5~7 d的Sprague-Dawley(SD)大鼠按照随机数字表法随机分为对照组、七氟醚全麻1组(S1组)和七氟醚全麻2组(S2组),每组8只。大鼠分娩后每组随机抽取8只仔鼠,维持生存30 d后,通过Morris水迷宫实验评估仔鼠学习记忆能力。收集出生后30 d的仔鼠脑标本,通过苏木精/伊红染色、尼氏染色观察海马和顶叶皮质神经元结构的形态变化和尼氏体。通过免疫组化法和蛋白质印迹法检测仔鼠海马和顶叶皮质中AIM2、CD45、白细胞介素1β(IL-1β)、pro-caspase-1和caspase-1 p10表达。结果 与对照组相比,S1组和S2组仔鼠长期学习和记忆能力较差,对海马和顶叶皮质的损伤程度不同。与对照组相比,S1组和S2组仔鼠中的AIM2、CD45、IL-1β、pro-caspase-1和caspase-1 p10表达升高(P均<0.05),且S2组高于S1组(P均<0.05)。结论 妊娠早期七氟醚全麻可促进仔鼠海马和顶叶皮质中AIM2表达及炎性反应的发生。 Objective To explore the effects of sevoflurane general anesthesia in the early pregnancy on the expression of interferon-inducible protein(absent in melanoma 2,AIM2)in the hippocampus and parietal cortex and inflammatory response of rat offspring.Methods Twenty-four Sprague-Dawley(SD)rats with a gestational age of 5-7 days were randomly divided into three groups:the control group and the sevoflurane general anesthesia group 1(S1 group),and sevoflurane general anesthesia group 2(S2 group),with 8 in each group.After all rats gave birth,8 newborn mice were randomly selected from each group to maintain their survival for 30 days.Morris water maze experiment was used to evaluate their learning and memory ability.Brain specimens were collected from pups at 30 days after birth,and the morphological changes of hippocampal and parietal cortex neurons and Nissl bodies were observed by hematoxylin/eosin staining and Nissl staining.The expression levels of AIM2,CD45,Interleukin-1β(IL-1β),pro-caspase-1 and caspase-1 p10 in the hippocampus and parietal cortex of ratoffsprings were detected by immunohistochemistry and Western blotting.Results Compared with the control group,the offsprings of the S1 and S2 groups had poorer long-term learning and memory abilities,and had different levels of damage to the hippocampus and parietal cortex.Compared with the control group,the expression levels of AIM2,CD45,IL-1β,pro-caspase-1 and caspase-1 p10 in the offsprings of the S1 and S2 groups increased(all P<0.05),and those were higher in the S2 group than in the S1 group(all P<0.05).Conclusion General anesthesia with sevoflurane in the first trimester promotes the expression of AIM2 in the hippocampus and parietal cortex of rat offsprings and the occurrence of inflammatory reactions.
