摘要
本研究对疑似禽腺病毒感染的鸡病料进行了病毒分离、PCR扩增基因与序列分析以及动物回归试验。结果显示:分离的病毒为球形、无囊膜,直径在70~80 nm,不具有凝集红细胞能力;毒株基因组全长43 634 bp,与Ⅰ群禽腺病毒同源性介于95.8%~99.7%,与Ⅱ群和Ⅲ群禽腺病毒同源性为34.4%~37.3%;毒株对3周龄SPF鸡的致死率为100%,死亡鸡可见典型的心包积液与包涵体肝炎症状,肝细胞染色可见嗜碱性包涵体。结果表明,本研究成功分离到1株Ⅰ群禽腺病毒C基因型血清4型病毒,命名为FAdV-4 SN2016,为下一步疫苗研制提供了坚实的物质基础。
Viral isolation and identification was performed using the materials collected from sick chicken suspect with the infection of Fowl adenoviruses.A serotype 4 Fowl adenovirus designated as FAdV-4 SN2016 was isolated through inoculation of the liver homogenate from a dead chicken into embryo and identified by electronic microscopy,PCR and sequencing.The virus particles were observed as spherical shape,icosahedral symmetry and non-enveloped with about 70~80 nm in diameter.The virus did not agglutinate red blood cells.The full-length genome of the SN2016 strain was 43634 bp by gene sequencing.It belonged to an evolutionary branch with reference strains of FAdV-4 as their nucleotide homology ranged from 95.8% to 99.7%.The isolated virus FAdV-4 SN2016 had nucleotide homology of 34.4% and 37.3% with group Ⅱ and group Ⅲ Fowl adenovirus.Animal experiment demonstrated that the infection of SN2016 virus via muscle inoculation into 3-week-age chickens result in typical inclusion body hepatitis and hydropericardium syndrome with 100% mortality.The basophilic inclusion bodies were found in liver cells from the sick chickens through HE staining.The study indicated that this highly pathogenic serotype 4 fowl adenovirus belonged to FAdV-C.The availability of the isolated virus FAdV-4 SN2016 would provide useful materials for further studies on the vaccine research.
