金钗石斛总生物碱脂质体对胃癌SGC-7901细胞的影响 被引量:1

Effects of Dendrobium nobile Lindl.alkaloids lipidosome on the proliferation, apoptosis and invasion of gastric cancer SGC-7901 cells

摘要 目的探讨金钗石斛总生物碱脂质体对胃癌SGC-7901细胞增殖、凋亡和侵袭的影响及其作用机制。方法分别用0.2、0.4、0.6、0.8和1.0 mg/mL金钗石斛总生物碱脂质体作用于SGC-7901细胞24、48和72 h,采用MTT检测细胞增殖情况,流式细胞计数法检测细胞凋亡情况,Transwell检测细胞侵袭情况,蛋白质印迹法检测凋亡相关蛋白Bcl-2、Bax和Caspase-3表达水平。结果采用不同浓度金钗石斛总生物碱脂质体作用于SGC-7901细胞后,细胞的增殖抑制率增加,差异均有统计学意义,均P<0.05。0.2、0.4和0.6 mg/mL金钗石斛总生物碱脂质体作用于SGC-7901细胞48 h后,细胞凋亡率分别为(6.23±0.60)%、(15.00±2.31)%和(24.10±1.50)%,对照组为(2.87±0.55)%,差异有统计学意义,均P<0.05;细胞侵袭数分别为44±4、32±5和22±4,对照组为57±6,差异有统计学意义,均P<0.05;凋亡相关蛋白Bax和Caspase-3表达水平明显升高(均P<0.05),Bcl-2表达水平明显降低,P<0.05。结论金钗石斛总生物碱脂质体能够明显抑制胃癌SGC-7901细胞增殖、促进细胞凋亡和抑制细胞侵袭,其作用机制可能与Bcl-2/Bax/Caspase-3信号通路有关,金钗石斛总生物碱脂质体可能成为胃癌治疗的药物。 Objective To explore the effects of Dendrobium nobile Lindl.alkaloids lipidosome on the proliferation, apoptosis and invasion of gastric cancer SGC-7901 cells and its mechanism.Methods SGC-7901 cells were treated with 0.2 mg/mL, 0.4 mg/mL, 0.6 mg/mL, 0.8 mg/mL and 1.0 mg/mL Dendrobium nobile Lindl.alkaloids lipidosome for 24 h, 48 h and 72 h, and MTT was used to detect cell proliferation;Flow cytometry was used to detect cell apoptosis;Transwell was used to detect cell invasion Situation;Western blot was used to detect the expression level of apoptosis-related protein Bcl-2, Bax and caspase-3. Results After treating SGC-7901 cells with different concentrations of Dendrobium nobile Lindl.alkaloids lipidosome, the cell proliferation inhibition rate increased significantly(P<0.05). Compared with the control group, after treating SGC-7901 cells with 0.2 mg/mL, 0.4 mg/mL and 0.6 mg/mL Dendrobium nobile Lindl.alkaloids lipidosome for 48 hours, the apoptosis rate was significantly increased [(2.87±0.55)% vs(6.23±0.60)%,(15.00±2.31)%,(24.10±1.50)%, P<0.05], the number of cell invasion was significantly reduced(57±6 vs 44±4,32±5,22±4, P<0.05), the expression level of apoptosis-related protein Bax and Caspase-3 was significantly increased(P<0.05), and the expression level of Bcl-2 was significantly decreased(P<0.05). Conclusions The Dendrobium nobile Lindl.alkaloids lipidosome can obviously inhibit the proliferation of gastric cancer SGC-7901 cells, promote cell apoptosis and inhibit cell invasion, and its mechanism may be related to Bcl-2/Bax/Caspase-3 signaling pathway. Dendrobium nobile Lindl.alkaloids lipidosome may be used as drugs for the treatment of gastric cancer.
