S100A2、Blimp1在多发性骨髓瘤患者骨髓单个核细胞中的表达水平及意义 被引量:1

Expression and significance of S100A2 and Blimp1 in bone marrow mononuclear cells of patients with multiple myeloma

摘要 目的研究S100钙结合蛋白(S100A2)、B淋巴细胞诱导成熟蛋白-1(Blimp1)在多发性骨髓瘤(MM)患者骨髓单个核细胞(BMMNCS)中的表达水平及意义。方法选取2018年3月至2020年5月于河南省中医院治疗的MM患者82例作为观察组(采用VAD治疗方案),同时选取同期贫血患者(常规贫血治疗)35例作为对照组。分别采用酶联免疫吸附法与反转录聚合酶链式反应法检测MM患者BMMNC中SS100A2、Blimp1的水平。观察比较两组一般资料及S100A2、Blimp1表达水平;采用Pearson相关性检验分析MM患者Blimp1、S100A2表达水平的相关性;建立受试者工作特征(ROC)曲线分析MM发生的预测价值。结果观察组S100A2表达水平显著低于对照组,差异有统计学意义(P<0.05);Blimp1表达水平显著高于对照,组差异有统计学意义(P<0.05)。S100A2、Blimp1表达水平均为MM发生的独立危险因素(P<0.05)。S100A2与Blimp1呈显著负相关(P<0.05)。S100A2、Blimp1及联合指标预测MM发生的曲线下面积(AUC)分别为0.881、0.836、0.945,联合指标预测价值更高,敏感度和特异度分别为97.1%与78.0%。结论S100A2、Blimp1表达水平均可作为MM发生的独立危险因素,且S100A2、Blimp1表达水平均可作为MM发生的预测因子,其中联合指标预测价值更高。 Objective To study the expression level and significance of S100 calcium binding protein(S100 A2)and B lymphocyte-induced mature protein-1(Blimp1)in bone marrow mononuclear cells(BMMNC)of patients with multiple myeloma(MM).Methods A total of 82 patients with MM who were treated in Henan Provincial Hospital of Traditional Chinese Medicine from March 2018 to May 2020 were selected as the observation group(using the VAD treatment plan),and 35 patients with anemia during the same period(conventional anemia treatment)were selected as the control group.Enzyme-linked immunosorbent assay and reverse transcription polymerase chain reaction were used to detect the levels of S100 A2 and Blimp1 in BMMNC of MM patients.The general data and the expression levels of S100 A2 and Blimp1 between the two groups were observed and compared.Pearson correlation test was used to analyze the correlation between the expression levels of Blimp1 and S100 A2 in MM patients.The receiver operating characteristic(ROC)curve was established to analyze the predictive value of the occurrence of MM.Results After univariate analysis,the expression level of S100 A2 in the observation group was significantly lower than that in the control group(P<0.05).The expression level of Blimp1 was significantly higher than that in the control group(P<0.05).The expression levels of S100 A2 and Blimp1 were independent risk factors for the occurrence of MM(P<0.05).S100 A2 was significantly negatively correlated with Blimp1(P<0.05).The area under the curve(AUC)of S100 A2,Blimp1 and the combination of the two predicting the occurrence of MM are 0.881,0.836,0.945,respectively.The combined detection has a higher predictive value,and the sensitivity and specificity are 97.1%and 78.0%,respectively.Conclusion The expression levels of S100 A2 and Blimp1 can be used as independent risk factors and predictors for the occurrence of MM,and the combination of the two has a higher predictive value.
