摘要
目的:探讨补气益精汤对糖尿病肾病模型大鼠肿瘤坏死因子-α和细胞间黏附因子-1的影响。方法:链脲佐菌素腹腔注射复制糖尿病肾病大鼠模型,随机将大鼠分为对照组、模型组,以及补气益精汤高、中、低剂量组。酶联免疫吸附试验法检测细胞间黏附因子-1、肿瘤坏死因子-α、白细胞介素-1β的表达情况,检测大鼠一般情况、氧化应激、肾功能指标。结果:与对照组比较,模型组大鼠肾脏细胞间黏附因子-1,白细胞介素-1β,肿瘤坏死因子-α的浓度均升高(P<0.05)。与对照组比较,模型组大鼠血糖、血肌酐、尿素氮、丙二醛、24小时尿蛋白明显升高,超氧化物歧化酶及过氧化氢酶降低(P<0.05);与模型组相比,补气益精汤组大鼠肾脏细胞间黏附因子-1、白细胞介素-1β、肿瘤坏死因子-α的浓度均下降(P<0.05),血糖、三酰甘油、胆固醇、血肌酐、尿素氮、丙二醛、24小时尿蛋白明显下降,超氧化物歧化酶及过氧化氢酶升高(P<0.05)。结论:补气益精汤可以降低糖尿病肾病模型大鼠细胞间黏附因子-1、肿瘤坏死因子-α及白细胞介素-1β的表达,有效改善肾脏功能。
Objective:To investigate the efficacy of the Buqi Yijing decoction(补气益精汤)on TNF-αand ICAM-1 in diabetic nephropathy model rats.Methods:The rat model of diabetic nephropathy was established by intraperitoneal injection of streptozotocin(STZ).The rats were divided into the control group,the model group,the Buqi Yijing decoction high-dose,medium-dose and low-dose groups by random number table method.The expressions of ICAM-1,TNF-αand IL-6 were detected by ELISA,and the general condition,oxidative stress and renal function indexes of rats were detected.Results:Compared with the control group,the concentrations of IL-6,ICAM-1,IL-1βand TNF-αin kidney in the model group were increased(P<0.05).Compared with the control group,blood glucose,serum creatinine,urea nitrogen,MDA,24-hour urinary protein in the model group were significantly increased(P<0.05),SOD and CAT were decreased(P<0.05).Compared with model group,the concentrations of ICAM-1,IL-1βand TNF-αin kidney in the Buqi Yijing decoction groups were decreased(P<0.05),the levels of blood glucose,triglycerides,cholesterol,serum creatinine,urea nitrogen,MDA,24-hour urinary protein were significantly decreased,and SOD and CAT were increased(P<0.05).Conclusion:The Buqi Yijing decoction can reduce the expression of ICAM-1 and TNF-αin diabetic nephropathy model rats,and effectively improve renal function.
