FBLIM1在人脑胶质瘤组织中的表达及其临床意义 被引量:2

摘要 目的探讨FBLIM1在人脑胶质瘤组织与正常脑组织表达水平的差异,以及其表达水平与胶质瘤病理分级、IDH1分子分型和患者预后的关系。方法从公共数据库中获取胶质瘤的数据,分析不同级别、IDH1分型胶质瘤组织的FBLIM1 mRNA表达水平及其与患者预后的关系。采用实时定量PCR、蛋白免疫印迹、免疫组织化学染色方法,检测临床手术切除的脑胶质瘤和脑外伤非瘤脑组织的FBLIM1表达水平。用Kaplan-Meier分析FBLIM1表达水平与患者总生存期(OS)的关系。结果公共数据库的胶质瘤数据分析和手术切除的胶质瘤标本检测均显示,FBLIM1表达水平随着胶质瘤病理分级升高而增高,高级别胶质瘤的FBLIM1表达水平显著高于非瘤脑组织;IDH1野生型胶质瘤的FBLIM1表达水平显著高于IDH1突变型(P<0.05~0.001)。生存分析显示,FBLIM1高表达组胶质瘤患者的OS明显短于低表达组,FBLIM1高表达组IDH1野生型高级别胶质瘤患者的OS短于低表达组(P<0.05~0.005)。结论FBLIM1在高级别胶质瘤组织的表达水平显著增高,IDH1野生型胶质瘤的FBLIM1表达水平高于IDH1突变型;FBLIM1高表达的胶质瘤患者的预后更差。FBLIM1可能是一个脑胶质瘤病理诊断和预后评估的生物标志物。 Objective To investigate the difference of FBLIM1 expression between human glioma and normal brain tissue,and the relationship between FBLIM1 expression and glioma pathological grade,IDH1 molecular typing and patient prognosis.Methods The data of gliomas were obtained from the public database,and the expression level of FBLIM1 mRNA in different grades and IDH1 typing gliomas and its relationship with patient prognosis were analyzed.The expression levels of FBLIM1 in gliomas and non tumor brain tissues after brain injury were detected by real-time quantitative PCR,Western Blot and immunohistochemical staining.Kaplan Meier was used to analyze the relationship between FBLIM1 expression level and overall survival(OS).Results The analysis of glioma data in public database and the detection of surgically resected glioma samples showed that the expression level of FBLIM1 increased with the increase of glioma pathological grade,and the expression level of FBLIM1 in high-grade glioma was significantly higher than that in non tumor brain tissues;The expression level of FBLIM1 in IDH1 wild-type gliomas was significantly higher than that in IDH1 mutant gliomas(P<0.05-0.001).Survival analysis showed that the OS of glioma patients in FBLIM1 high expression group was significantly shorter than that in low expression group,and the OS of IDH1 wild-type high-grade glioma patients in FBLIM1 high expression group was shorter than that in low expression group(P<0.05~0.005).Conclusions The expression level of FBLIM1 in high-grade gliomas was significantly higher than that in IDH1 mutant gliomas;The prognosis of glioma patients with high expression of FBLIM1 is worse.FBLIM1 may be a biomarker for pathological diagnosis and prognostic evaluation of glioma.
