女贞子-黄芪抗胃癌研究及作用机制的探讨

Effect of ligustri lucidi fructus-astragalus radix against gastric cancer and its mechanism

目的女贞子-黄芪的抗胃癌效果验证及抗胃癌作用靶点探究。方法借助MTT实验验证女贞子-黄芪的抗胃癌效果。利用中药系统药理学数据库与分析平台(TCMSP)库,获取药对活性成分及作用靶标。利用UniProt数据库和Genecards数据库获取药对及胃癌靶标,通过STRING网站及Cytoscape 3.6.0构建蛋白作用网络以及“成分-靶标”网络。通过Kaplan-Meier plotter网站评估靶标基因与胃癌患者预后的关系。运用分子对接技术对女贞子-黄芪共有活性成分及中心靶标进行对接模拟。结果女贞子-黄芪抑制胃癌细胞的生长,50μg/ml浓度下,胃癌细胞的24 h、48 h、72 h的抑制率分别为(5.44±1.50)%、(19.23±2.29)%、(14.34±2.95)%,差异均有统计学意义。通过筛选得到女贞子-黄芪的31个活性成分和189个胃癌作用靶标,其核心靶标与共有活性成分均有良好的对接效果。结论女贞子-黄芪抑制了胃癌细胞的增殖,AKT1可能是女贞子-黄芪治疗胃癌的关键靶点。

Objective To verify the anti-gastric cancer(anti-GC)effect of ligustri lucidi fructus-astragalus radix and to explore its anti-GC targets.Methods MTT assay was used to verify the anti-GC effect of ligustri lucidi fructus-astragalus radix.Traditional Chinese Medicine Database and Analysis Platform(TCMSP)was used to obtain the active ingredients and targets of the drug pair.UniProt database and Genecards database were used to obtain the drug pair and GC targets.The protein action network and"component-target"network were constructed through string website and Cytoscape 3.6.0.The relationship between target genes and the prognosis of GC patients was evaluated through the Kaplan-Meier plotter website.The molecular docking technology was used to perform the docking simulation of the common active ingredients and central targets of ligustri lucidi fructus-astragalus radix.Results Ligustri lucidi fructus-astragalus radix inhibited the growth of GC cells.At a concentration of 50μg/ml,the inhibition rates of GC cells at 24 h,48 h and 72 h were(5.44±1.50)%,(19.23±2.29)%and(14.34±2.95)%,respectively,with statistical differences.Through screening,31 active ingredients of ligustri lucidi fructus-astragalus radix and 189 GC targets were obtained.The core targets and common active ingredients showed good docking effects.Conclusion Ligustri lucidi fructus-astragalus radix inhibits the proliferation of GC cells.AKT1 may be the key target of ligustri lucidi fructus-astragalus radix in the treatment of GC.
