银黄颗粒超高效液相色谱指纹图谱研究

摘要 研究建立了银黄颗粒超高效液相色谱(UPLC)指纹图谱测定方法,应用相似度法、聚类分析法、主成分分析法分析样品间的异同,为其鉴别与质量控制提供了参考。采用DAD-UPLC,ZORBAX SB-C18(Φ3.0 mm×100 mm,1.8μm)色谱柱,以0.05%磷酸水溶液和乙腈为流动相,梯度洗脱,检测波长为318 nm,建立了银黄颗粒的指纹图谱,指纹图谱中有12个共有峰,20批次样品指纹图谱相似度在0.982~0.999,聚类分析和主成分分析可以看出不同厂家产品间是存在一定的差异的。该研究建立的UPLC指纹图谱有较好的精密度、重复性和稳定性,相似度分析能够快速判断产品的一致性,聚类分析和主成分分析能够更加细致的分辨不同厂家产品间的差异。该方法适用于对银黄颗粒的质量进行客观评价。 Ultra Performance Liquid Chromatography(UPLC)fingerprint of Yinhuang granules was established in this study.The similarities and differences among samples were analyzed by similarity method,cluster analysis method and principal component analysis method,which provided reference for their identification and quality control.The fingerprint of Yinhuang granules was established by using ZORBAX SB-C18(3.0 mm×100 mm,1.8μm)chromatographic column;0.05%phosphoric acid aqueous solution and acetonitrile as mobile phase;318 nm as detection wavelength.There were 12 common peaks in the UPLC fingerprint of Yinhuang granules.The fingerprint similarity of 20 batches of Yinhuang granules was above 0.98.Cluster analysis and principal component analysis show that there are some differences among Yinhuang granules from different manufacturers.This method is suitable for the objective evaluation of the quality of Yinhuang granules.
