去整合素-金属蛋白酶12(ADAM12)是去整合素-金属蛋白酶家族成员之一,参与了细胞-细胞、细胞-基质的一系列生物学过程。在多种肿瘤类型中,ADAM12表达量明显升高,促进了肿瘤的形成、增殖和转移等过程,与肿瘤分期、患者生存率、肿瘤浸润等多种临床表型密切相关。同时,血清中ADAM12表达量与肿瘤分期相关,具有肿瘤无创诊断标志物的潜能。该文就ADAM12在多种肿瘤中的功能和分子机制进行综述。

A deintegrin-metalloproteinase 12(ADAM12)is a member of a deintegrin and metalloproteinase family,which has been implicated in a variety of biological processes involving cell-cell and cell-matrix interactions.High expression of ADAM12 in multiple types of solid tumors promote the tumorigenesis process,including cancer formation,proliferation and invasion,and significantly associate with various clinical phenotype(e.g.tumor grade,patient survival and tumor invasion).Meanwhile,serum levels of ADAM12 correlate with tumor grade and stage,indicating that ADAM12 may have potential of no-invasive diagnostic biomarker.This review focus on the function and molecular mechanism of ADAM12 across various types of solid tumors.
