摘要
DEAD-box RNA解旋酶3(DDX3)属于DEAD-box蛋白家族的一员,具有依赖ATP酶的解旋酶活性,其功能是调控RNA代谢的各个方面,包括翻译、转录、RNA稳定性、mRNA输出和mRNA剪接,进一步参与肿瘤增殖、肿瘤侵袭和转移、肿瘤耐药和肿瘤免疫治疗等多种过程。DDX3在人类癌症中的表达和功能具有组织特异性,在不同肿瘤甚至同一种肿瘤中的作用都具有争议。本文就DDX3在肿瘤中的研究进展及功能差异的相关原因展开综述。
Dead-box RNA helicase 3(DDX3),a member of the Dead-box protein family,has ATPasedependent helicase activity.It mainly plays a physiological role in cytoplasm and nucleus,and functions to regulate all aspects of RNA metabolism,including translation,transcription,RNA stability,mRNA output and mRNA splice.It is further involved in tumor proliferation,tumor invasion and metastasis,tumor drug resistance and tumor immunotherapy.The expression and function of DDX3 in human cancers are tissuespecific,and the role of DDX3 in different tumors or even in the same tumor has been controversial.This paper reviews the research progress of DDX3 in tumors and the related reasons of functional differences.
