生物工程综合实验“真菌α-淀粉酶固态发酵生产”的教学改革与实践 被引量:2

摘要 生物工程专业综合实验教学能够提升学生综合实验技能,以及培养学生解决问题的能力和创新意识。淮南师范学院生物工程综合实验以“真菌α-淀粉酶固态发酵生产”为研究对象,实验安排学生开展真菌α-淀粉酶产生菌株筛选、菌种分子生物学鉴定、菌株紫外线诱变选育、30L固态发酵罐实罐灭菌、固态发酵生产真菌α-淀粉酶、板框压滤结构分离浸提酶液、陶瓷膜浓缩酶液、喷雾干燥制备固体酶制剂、蛋白质凝胶层析纯化真菌α-淀粉酶蛋白、SDS-PAGE测定酶蛋白分子量大小等9个模块实验。该综合实验以连贯性的研究任务驱动学生开展探究性学习,实验以学生为主体,采用连续运行的方式安排教学任务,保证模块实验的整体性和连贯性。采用“考勤+课堂实验操作+平时实验报告+实践操作考试”的方式进行考核,强化学生对综合实验操作技能的掌握。实践证明,本综合实验的开设显著增强了学生对专业理论知识和实验技能的掌握程度,强化了学生解决实际问题能力,取得了较好的教学改革成效。 Bioengineering integrated experiment could help to improve the comprehensive experimental skills for bioengineering major students,as well as enhancing their ability of analyzing and solving problems,and activating their enthusiasm for creativity.The program of the bioengineering comprehensive experiment in our school is based on"solid-state fermentation of fungiα-amylase",and the bio-engineering major students have been assigned with 9 experimental units that include screening fungalα-amylase producing strains,identification of the obtained strains by molecular biology,mutation breeding of the strains by ultraviolet radiation,sterilization of 30L solid-state fermentation tank,solid-state fermentation of fungiα-amylase in 30L fermentation tank,solid-liquid separation of fungiα-amylase by plate and frame pressure filtration,incrassation of fungiα-amylase solution by ceramic membrane,preparation of solid enzyme preparation by spray drying,purification of fungiα-amylase by Sephadex G50,and measuring of the molecular weight of fungiα-amylase protein by SDS-PAGE.This experimental research module will guide students to experience an entire inquiry-based learning process by consistently planning and conducting research projects,The comprehensive experiment also helps to enhance the connection between practical teaching and theoretical courses of bioengineering specialty.The experimental module is student-centered,implemented with"continuously operating"mode,to ensure its integrity and consistency.The integrated evaluation of"attendance","classroom experimental operation","usual experiment report",and"practical operation examination"were adopted in the bioengineering experiment to help the bioengineering major students easily master the comprehensive experimental operation skills.It is proved in practice that teaching reform has helped to enhance the students'understanding mastery of professional theoretical knowledge and experimental skills,strengthen students'ability to solve practical problems,and achieved good results in teaching reform.
