超高效液相色谱-串联四级杆质谱法测定中草药制剂中非法添加的糖皮质激素和抗真菌药物

Determination of Glucocorticoids and Antifungal Drugs in Chinese Herbal Medicine by UPLC-MS/MS

目的:建立中草药制剂中41种糖皮质激素和8种抗真菌药物的超高效液相色谱-串联四级杆质谱(UPLC-MS/MS)测定方法。方法:色谱柱为Kinetex-C18(4.6 mm×50 mm,2.6 μm),梯度洗脱:流动相A为一级水,流动相B为乙腈,流速0.3 mL/min。正离子扫描,扫描方式MRM。结果:该方法有良好的线性关系,相关系数r>0.990 0,在典型的中草药制剂中,不同添加水平下方法的回收率范围为88.3%~96.1%,通过该法检出中草药制剂阳性样品3个。结论:该方法简便、快速、准确,可用于41种糖皮质激素和8种抗真菌药物的测定。

Objective:To establish an UPLC-MS/MS method for the determination of 41 glucocorticoids and 8 antifungal drugs in Chinese herbal medicine preparations.Method:A Kinetex-C18 (4.6 mm×50 mm,2.6 μm) column was used the gradient elution of water and acetonitrile as the mobile phase and the flow rate was 0.3 mL/min.The electrospray ionization source in positiveion mode was used for multiple reaction monitoring mode.Results:The method presented a good linear relation (r>0.990 0).In typical Chinese herbal medicine preparations,the recovery range of the method was 88.3%-96.1% at different levels.Three positive samples of chinese herbal medicine preparations were successfully detected by this method.Conclusion:The method was convenient,sensitive and accurate,it could be used for determination of 41 glucocorticoids and 8 antifungal drugs in Chinese herbal medicine preparations.
