响应面-满意度函数法优化绵马贯众中多糖及黄酮类成分的共提取工艺 被引量:1

Optimization of co-extraction of polysaccharides and flavonoids from Dryopteris crassirhizoma by Response surface methodology-satisfaction function

摘要 目的利用响应面-满意度函数法优化绵马贯众多糖及黄酮类成分的共提取工艺。方法在单因素实验的基础上,以乙醇浓度、料液比、超声时间和超声温度为自变量,以葡萄糖和芦丁为对照,以绵马贯众黄酮和多糖总得率的满意度为响应值,应用中心组合设计实验方法,研究各自变量及交互作用对满意度的影响,建立二次多项回归方程的数学模型。结果各因素影响的大小程度为:料液比>乙醇浓度>超声温度>超声时间,且各因素的一次项和二次项对绵马贯众多糖及黄酮得率的满意度影响均比较显著。最优共提条件为:料液比1:25 g·mL^(-1),30%乙醇于49℃超声25 min;此时满意度为0.731,与预测值相差极小。结论优化后的提取工艺可靠、方法可行,可用于绵马贯众多糖及黄酮成分的共提取。 OBJECTIVE To optimize the process conditions of polysaccharides and flavonoids from Dryopteris crassirhizoma NaKai by Response surface methodology-satisfaction function.METHODS On the basis of single factor experiment, the ethanol concentration, ratio of solid to liquid, ultrasonic time and ultrasonic temperature were taken as independent variables, with glucose and rutin as control and satisfaction degree of the yield of flavone and polysaccharide from D.crassirhizoma as response value, the effect of each variation and its interaction on the satisfaction degree of flavone and polysaccharide was studied by using the central composite design method.RESULTS The influence degree of each factor was material-liquid ratio>ethanol concentration>ultrasonic temperature>ultrasonic time, and the first and second terms of each factor had significant effect on the content of co-extraction of polysaccharide and flavone from D.Crassirhizoma.The optimal co-extraction conditions were the ratio of solid to liquid of 1:25 g·mL;,the concentration of ethanol of 30%,the ultrasonic time of 25 min, the ultrasonic temperature of 49 ℃ and the satisfactory degree of 0.731,resulting in that the difference was very small.CONCLUSION The developed extraction process appears reliable and feasible, and can be used for the extraction of polysaccharide and flavone from D.crassirhizoma.
