响应面法优化红菊苣花青素的提取工艺及其微胶囊的制备 被引量:5

摘要 目的采用响应面法优化红菊苣花青素的提取工艺,并利用提取得到的红菊苣花青素制备微胶囊。方法应用单因素试验和响应面法优化超声波辅助双水相提取红菊苣花青素的工艺,利用响应面设计模型对乙醇质量分数、硫酸铵质量浓度、液料比、超声波频率等提取参数进行了优化,选取最优条件。并进一步对提取得到的红菊苣花青素进行微胶囊化,采用红外光谱、扫描电镜对微胶囊化颗粒进行分析。结果超声波辅助双水相提取红菊苣花青素最佳工艺条件为:乙醇质量分数为26%,硫酸铵质量分数为22%,超声频率为36 kHz,液料比为26:1 (mL/g),提取花青素含量可达到最高值,为14.33μg/g。微胶囊中芯材与壁材包埋成功,且微胶囊呈球状、表面光滑。结论优化的提取工艺稳定、可靠,微胶囊中芯材与壁材包埋成功,具有很好的实际应用价值。 Objective To optimize the extraction process of anthocyanins of from Cichorium intybus by response surface methodology,and prepare microcapsules from the extracted anthocyanins from Cichorium intybus.Methods The ultrasonic assisted aqueous two-phase extraction process of anthocyanins of from Cichorium intybus was optimized by single factor test and response surface methodology.The extraction parameters such as ethanol mass fraction,ammonium sulfate mass concentration,liquid-solid ratio and ultrasonic frequency were optimized by response surface design model,and the optimal conditions were selected.The extracted anthocyanin from Cichorium intybus was further microencapsulated,and the microencapsulated particles were analyzed by infrared spectroscopy and scanning electron microscopy.Results The optimum conditions of ultrasonic assisted aqueous two-phase extraction of anthocyanins from Cichorium intybus were as follows:Ethanol mass concentration 26%,ammonium sulfate mass concentration 22%,ultrasonic frequency 36 kHz and liquid-solid ratio 26:1(mL/g),on which condition the extract apricot content could reach the highest(14.33 μg/g).The core material and wall material in the microcapsule were successfully embedded,and the microcapsule was spherical and the surface was smooth.Conclusions The optimized extraction process is stable and reliable,and the core and wall materials in microcapsules are successfully embedded,which has good practical value.
