外环境空气细颗粒物对COPD大鼠致病影响的研究

Effects of PM_(2.5) on the pathogenicity of chronic obstructive pulmonary disease in rats

摘要 目的研究外环境空气细颗粒物(PM2.5)是否会对慢性阻塞性肺疾病(COPD)大鼠造成致病影响。方法清洁级大鼠购自河北医科大学动物实验室。18只大鼠COPD造模成功后随机分为3组, 每组6只, COPD大鼠吸入清洁空气组(A组), COPD大鼠吸入PM2.5暴露2周组(B组), COPD大鼠吸入PM2.5暴露4周组(C组)。对实验大鼠进行肺功能评估, 肺部炎症因子检测及基因表达差异分析, 探讨外环境空气PM2.5对COPD大鼠致病性的影响。结果 PM2.5的暴露可以引起COPD大鼠肺部组织的炎症反应增加。随着暴露时间的延长, 炎症反应呈加重趋势, 会导致COPD大鼠肺功能进行性下降, 肺功能受损。COPD大鼠经PM2.5的暴露后, 基因表达受到影响, 表达上调基因328个, 表达下调基因81个。表达差异的基因与COPD疾病的发生、发展及病程恶化相关, 与肺部组织的慢性持续炎症及结构重塑相关。结论 PM2.5的暴露可以引起COPD大鼠肺部组织的炎症反应加重, 且随着暴露时间的延长, 炎症反应呈加重趋势, 肺功能呈恶化趋势。PM2.5的暴露会诱导大鼠基因表达变化。持续PM2.5的暴露, 会诱发气道的长期持续慢性炎症反应和气道的结构重塑, 诱发COPD的进行性进展。 Objective To study the pathogenic effects of ambient air fine particulate matter(PM2.5)on chronic obstructive pulmonary disease(COPD)in rats.Methods Clean rats were purchased from animal laboratory of Hebei Medical University.After successful modeling of COPD,the 18 rats were randomly divided into three groups,with six in each group.COPD rats exposed to clean air(group A),COPD rats exposed to PM2.5 for 2 weeks(group B),and COPD rats exposed to PM2.5 for 4 weeks(group C).Lung function was evaluated,lung inflammatory factors detected,and gene expression differences analyzed to explore the influence of ambient air PM2.5 on the pathogenicity of COPD rats.Results Exposure to PM2.5 can cause an increased inflammatory response in lung tissues of COPD rats.With the extension of exposure time,inflammatory response tends to worsen,leading to progressive decline and impaired lung function in COPD rats.After exposure to PM2.5,gene expression in COPD rats was affected,with 328 genes up-regulated and 81 down-regulated.Differentially expressed genes are associated with the occurrence,progression,and deterioration of COPD disease,as well as chronic persistent inflammation and structural remodeling of lung tissues.Conclusions Exposure to PM2.5 can aggravate the inflammatory response in the lung tissues of COPD rats.With the extension of exposure time,the inflammatory response tends to worsen and lung function tends to deteriorate.Exposure to PM2.5 induces changes in gene expression in rats.Continuous exposure to PM2.5 can induce long-term chronic inflammatory response and airway remodeling,and induce the progressive development of COPD.
