男性双相情感障碍患者外周血相关基因表达与血清细胞因子水平的相关性研究 被引量:3

A Study on the Correlation Between the Expression of Peripheral Blood Related Genes and the Level of Inflammatory Cytokines in Male Patients with Bipolar Disorder

摘要 目的研究男性双相情感障碍(bipolar disorder,BD)患者外周血谷氨酸受体亚单位-1(GluD1)、突触结合蛋白7(Syt7)、M2型瞬时受体电位(TRPM2)以及甲壳质酶蛋白40(YKL-40)基因表达变化,并探讨与血清炎性细胞因子水平的相关性。方法选择我院2017年5月至2019年5月收治的86例男性BD患者为观察组;同时期来我院健康男性体检者40例作为对照组。qRT-PCR法检测对照组和观察组患者外周血GluD1、YKL-40、Syt7、TRPM2基因的表达水平;酶联免疫吸附法检测血清炎性细胞因子IL-1、IL-4、IL-6、IL-10以及TNF-α的水平,并对相关基因表达水平与炎性细胞因子水平的关系进行分析。结果与对照组比较,观察组基因GluD1和Syt7的表达水平明显降低,而TRPM2和YKL-40的表达水平明显升高(P<0.05),血清炎性细胞因子IL-1、IL-4以及TNF-α水平显著升高(P<0.05),而IL-6及IL-10水平差异无统计学意义(P>0.05)。相关性分析结果显示,GluD1基因水平与IL-1和TNF-α水平呈负相关(r=-0.251,-0.200;均P<0.05),Syt7基因水平与IL-1水平呈负相关(r=-0.186,P<0.05),TRPM2基因水平与IL-6水平呈正相关(r=0.190,P<0.05),YKL-40基因水平与IL-1和IL-6水平呈正相关(r=0.187,0.220;均P<0.05)。结论GluD1、Syt7、TRPM2以及YKL-40基因在男性BD患者外周血中差异表达,且与不同的血清炎性细胞因子水平相关。 Objective To study the expression change of glutamate receptor subunit-1(GluD1),synaptic binding protein 7(Syt7),type M2 transient receptor potential(TRPM2)and chitinase protein 40(YKL-40)genes in peripheral blood of male patients with bipolar disorder(BD),and to explore their correlation with serum inflammatory cytokine levels.Methods A total of 86 male BD patients admitted to our hospital from May,2017 to May,2019 were selected as the observation group,while 40 healthy male physical examinees during the same period as the control group.The expression levels of GLUD1,YKL-40,Syt7 and TRPM2 genes in the peripheral blood of the control group and the observation group were measured by qRT-PCR assay.The enzyme-linked immunosorbent assay was conducted to detect levels of serum inflammatory cytokines IL-1,IL-4,IL-6,IL-10 and TNF-α.And the relationship between the expression levels of related genes and the levels of inflammatory cytokines were analyzed.Results Compared with the control group,the expression levels of GluD1 and Syt7 genes in the observation group were significantly reduced,while the expression levels of TRPM2 and YKL-40 genes were significantly increased(P<0.05).Serum levels of inflammatory cytokines IL-1,IL-4 and TNF-αwere significantly increased(P<0.05),while the levels of IL-6 and IL-10 had no significantly difference(P>0.05).Correlation analysis results showed that GluD1 gene level was negatively correlated with IL-1 and TNF-αlevels(r=-0.251,-0.200;both P<0.05),Syt7 gene level was negatively correlated with IL-1 level(r=-0.186,P<0.05),TRPM2 gene level was positively correlated with IL-6 level(r=0.190,P<0.05),and YKL-40 gene level was positively correlated with IL-1 and IL-6 levels(r=0.187,0.220;both P<0.05).Conclusion The expressions of GluD1,Syt7,TRPM2 and YKL-40 genes are different in the peripheral blood of male BD patients,and correlate with different levels of serum inflammatory cytokines.
