摘要
目的:探究红景天苷对烟雾暴露大鼠肺微血管内皮细胞(PMVEC)活性及细胞核因子-κB(NF-κB)/Bcl-2表达水平的影响。方法:将大鼠随机分为空白组,烟雾组,红景天苷低、中、高剂量组。体外培养大鼠PMVEC,MTT法检测各组PMVEC活性,流式细胞仪检测各组PMVEC凋亡率,H-E染色观察各组大鼠肺病理组织学改变,免疫印迹法检测各组大鼠肺组织NF-κB和Bcl-2蛋白表达。结果:H-E染色结果显示,和空白组比较,烟雾组大鼠可见支气管壁和肺泡结构的破坏,支气管管腔结构变窄,炎症细胞浸润,有黏性液分泌物,肺泡腔变大、壁薄;肺泡间隔变窄或中断,还可见肺大泡;红景天苷低、高剂量组的肺组织结构出现明显改善,且红景天苷高剂量组的肺组织改善最为明显。MTT结果显示,烟雾组大鼠24 h和48 h的PMVEC活性明显低于空白组;和烟雾组比较,红景天苷低、中、高剂量组大鼠的PMVEC活性显著提升,且红景天苷高剂量组大鼠PMVEC活性升高最为显著。细胞凋亡结果显示,和空白组比较,烟雾组PMVEC凋亡率显著增加,红景天苷低、中、高剂量组PMVEC凋亡率显著降低,且随着红景天苷剂量的增加,凋亡率随之降低。和空白组大鼠比较,烟雾组NF-κB蛋白表达显著升高,Bcl-2蛋白表达降低,红景天苷低、中、高剂量组NF-κB蛋白表达显著低于烟雾组,Bcl-2蛋白表达显著升高,且红景天苷高剂量组的效果最为显著。结论:红景天苷可抑制烟雾暴露大鼠PMVEC的凋亡,其作用机制可能与抑制NF-κB的活性、促进Bcl-2表达有关。
Objective:To investigate the effect of salidroside on the activity of pulmonary microvascular endothelial cell(PMVEC)and the expression of nuclear factor-κB(NF-κB)/Bcl-2 in smoke exposed rats.Methods:The rats were randomly divided into blank group,smoke group,low-dose group,medium-dose and high-dose salidroside group.PMVEC was cultured in vitro.The activity of PMVEC was detected by MTT assay,and the apoptosis rate of PMVEC was detected by flow cytometry.H-E staining was used to observe the pathological changes of lung tissues in each group.The protein expression of NF-κB and Bcl-2 in lung tissues of rats in each group was detected by Western blotting.Results:H-E staining results showed that compared with blank group,the bronchial wall and alveolar structure were damaged,and the bronchial lumen structure was narrowed in smoke group.Inflammatory cell infiltration,mucous fluid secretion,enlarged alveolar cavity,and thin wall were observed.Alveolar septa narrowed or interrupted,also visible pulmonary bullae appeared.The lung tissue structure of the three salidroside groups was significantly improved,and the lung tissue of salidroside high-dose group was the most significantly improved.MTT results showed that PMVEC activity at 24 h and 48 h in smoke group was significantly lower than that in blank group.Compared with smoke group,PMVEC activity of the three salidroside groups was significantly increased,and PMVEC activity of salidroside high-dose group was the most significant increased.Apoptosis results showed that compared with blank group,the apoptosis rate of PMVEC in smoke group was significantly increased,and the apoptosis rate of PMVEC in the three salidroside groups was significantly decreased,and the apoptosis rate decreased with the increase of salidroside dose.Compared with blank group,NF-κB protein expression was increased and Bcl-2 protein expression was decreased in smoke group.NF-κB protein expression in the three salidroside groups was significantly lower than that in smoke group,and Bcl-2 protein expression was significantly increased in salidroside high dose group,and the effect of salidroside high dose group was most significant.Conclusion:Salidroside can inhibit the apoptosis of PMVEC cells in smoke-exposed rats,and its mechanism may be related to inhibiting the activity of NF-κB and promoting the expression of Bcl-2.
