高地钩叶藤材薄壁细胞壁主要化学成分的微区分布

摘要 为构建棕榈藤材薄壁细胞壁结构模型,扩大棕榈藤材的用途、提高棕榈藤材的利用率,以高地钩叶藤为研究对象,在藤茎高2 m处截取试样,经软化、聚乙二醇包埋、切片、硼氢化钠(NaBH_(4))溶液浸泡、显微共聚焦拉曼光谱仪逐点扫描、获取光谱数据、分析。研究结果表明:薄壁细胞的次生壁(S)上纤维素的集团数量比最高,复合胞间层(CML)的纤维素集团数量比次之,而细胞角隅(CC)处纤维素的集团数量比最低;次生壁外部(SⅠ)、中部(SⅡ)和内部(SⅢ)3个形态区纤维素的集团数量比差别较小;CML与SⅡ处纤维素集团数量比没有差别。CC处的木质素集团数量比最高,CML次之,S上的木质素集团数量比最低。SⅠ、SⅡ和SⅢ3个形态区木质素集团数量比差别较小,SⅡ木质素集团数量比小于CML。S层上的半纤维素集团数量比最高,CML次之,CC处集团数量比最低。薄壁细胞壁上纤维素、半纤维素集团数量比由高至低依次是S、CML和CC,而木质素集团数量比分布相反。SⅠ、SⅡ和SⅢ3个形态区域纤维素集团数量比、木质素集团数量比差别均较小。 In order to construct the structure model of parenchyma cell wall,expand the use and improve the utilization rate of rattan,Plectocomia himalayana was chosen as the research object,and from which the samples were selected at a height of 2 m from the ground.After being softened,embedded with polyethylene glycol,sliced,slices being soaked in NaBH_(4),the spectral data was obtained by point-by-point scanning with the Microscopic Confocal Raman Spectrometer and analyzed.The highest concentration of cellulose was at the secondary wall(S),followed by the composite intercellular layer(CML),and the lowest at the cell corner(CC).There was little difference between the cellulose contents in the outer(SⅠ),middle(SⅡ)and inner(SⅢ)of the secondary wall,and there was no difference between CML and SⅡ.The lignin concentration at CC was the highest,followed by CML,and the S was the lowest.There was also no significantly difference among SⅠ,SⅡand SⅢ,and the lignin content at SⅡwas less than CML.The hemicellulose content in S layer was the highest,followed by CML and lowest at CC.The contents of cellulose and hemicellulose gradually decreased in S,CML and CC,while the lignin content was opposite.There were no distinct differences of cellulose concentration and lignin concentration among SⅠ,SⅡand SⅢ.
