裸花紫珠乙酸乙酯提取物对结肠癌细胞增殖、迁移和侵袭的影响 被引量:1

摘要 目的探究裸花紫珠(CN)提取物调控10号染色体上缺失的磷酸酶和张力蛋白同源物(PTEN)对结肠癌细胞增殖、迁移、侵袭的影响。方法体外培养人结肠癌细胞SW480,随机分为对照组、CN乙酸乙酯100 mg/L组、CN乙酸乙酯150 mg/L组、CN乙酸乙酯200 mg/L组、CN乙酸乙酯200 mg/L+si-NC组、CN乙酸乙酯200 mg/L+si-PTEN组。MTT法、Transwell实验分别检测增殖、迁移及侵袭能力;western blot检测Cyclin D1、N-cadherin、p21、E-cadherin、PTEN蛋白表达量。结果与对照组比较,CN乙酸乙酯200 mg/L组细胞活力和Cyclin D1、N-cadherin蛋白水平降低,迁移及侵袭细胞数明显减少,而p21、E-cadherin、PTEN蛋白水平升高,差异均有统计学意义(P<0.05);与CN乙酸乙酯200 mg/L+si-NC组比较,CN乙酸乙酯200 mg/L+si-PTEN组细胞活力升高,Cyclin D1、N-cadherin蛋白水平升高,迁移及侵袭细胞数增多,PTEN、p21、E-cadherin蛋白水平降低,差异均有统计学意义(P<0.05)。结论裸花紫珠在人结肠癌细胞的增殖、迁移、侵袭中发挥重要作用,其可能通过调控PTEN的表达影响结肠癌细胞的增殖、迁移和侵袭。 Objective To explore the effect of ethyl acetate extract of Callicarpa nudiflora on the proliferation,migration,and invasion of colon cancer cells by regulating phosphatase and tensin homolog deleted on chromosome 10(PTEN).Methods Human colon cancer cells SW480 were cultured in vitro and randomly divided into groups:control group,ethyl acetate extract of Callicarpa nudiflora 100 mg/L group,ethyl acetate extract of Callicarpa nudiflora 150 mg/L group,ethyl acetate extract of Callicarpa nudiflora 200 mg/L group,ethyl acetate extract of Callicarpa nudiflora 200 mg/L+si-NC group,ethyl acetate extract of Callicarpa nudiflora 200 mg/L+si-PTEN group.MTT method and Transwell test were used to detect proliferation,migration and invasion.Western blot was used to detect the expression of Cyclin D1,N-cadherin,p21,E-cadherin,PTEN protein.Results Compared with the control group,the cell viability and the levels of Cyclin D1,N-cadherin of the ethyl acetate extract of Callicarpa nudiflora 200 mg/L group were decreased,number of migration and invasive cells were decreased,while the levels of p21,E-cadherin,PTEN were increased,all with statistically significant differences(P<0.05).Compared with the ethyl acetate extract of Callicarpa nudiflora 200 mg/L+si-NC group,the cell viability and the levels of Cyclin D1,N-cadherin in the ethyl acetate extract of Callicarpa nudiflora 200 mg/L+si-PTEN group were increased,number of migration and invasive cells were increased,the levels of PTEN,P21,E-cadherin were decreased,all with statistically significant differences(P<0.05).Conclusion Callicarpa nudiflora plays an important role in the proliferation,migration,and invasion of human colon cancer cells.It may affect the proliferation,migration,and invasion of colon cancer cells by regulating the expression of PTEN.
