贝莱斯芽孢杆菌抑尖孢镰刀菌脂肽类物质的鉴定 被引量:11

Identification of Fusarium oxysporum inhibitory peptides from Bacillus velezensis

摘要 为探究贝莱斯芽孢杆菌(Bacillus velezensis)P9对于尖孢镰刀菌(Fusarium oxysporum)具有拮抗作用的主要活性成分,采用酸沉淀法和有机溶剂沉淀法提取菌贝莱斯芽孢杆菌P9发酵液中的脂肽类抗生素粗提物,研究粗提物对尖孢镰刀菌的抑菌活性。利用多功能制备液相仪对其组分进行分离,确定具有抑菌活性的组分,并利用高效液相色谱-四级杆飞行时间质谱联用(HPLC-Q-TOF-MS)仪对具有抑菌活性的组分进行鉴定。结果表明,脂肽类抗生素粗提物对尖孢镰刀菌有良好的抑菌作用,且酸沉淀法得到的粗提物抑菌活性较高,从中共分离到7种组分(编号为1^(#)~7^(#)),其中4种组分具有抑菌作用,经鉴定,组分2^(#)为C_(15)SurfactinA或C_(16)SurfactinB或C_(15)SurfactinC,组分4^(#)、6^(#)、7^(#)分别为C_(15)BacillomycinD、C_(14)BacillomycinD、C_(16)BacillomycinD。 In order to explore the main active ingredient of Bacillus velezensis P9 which had an antagonistic effect on Fusarium oxysporum,the crude extract of lipopeptide antibiotics from the fermentation broth of B.velezia P9 was extracted by acid precipitation method and organic solvent precipitation method,and the antibacterial activity of the crude extract against F.oxysporum was studied.Then the components were separated using multifunctional preparation liquid phase meter,and the components with antibacterial activity were determined.Finally,the components with antibacterial activity were identified using high performance liquid chromatography-quadrupole time-of-flight mass spectrometry(HPLC-Q-TOF-MS).The results showed that the crude extract of lipopeptide antibiotics had good antibacterial effect on F.oxysporum,and the crude extract obtained by acid precipitation method had high antibacterial activity.There were 7 kinds of components isolated from crude extract(numbered 1^(#)-7^(#)),and 4 kinds of components had antibacterial effects.After identification,component 2^(#)was C_(15)SurfactinA or C_(16)SurfactinB or C_(15)Surfactin C,and components 4^(#),6^(#)and 7^(#)were C_(15)BacillomycinD,C_(14)BacillomycinD and C_(16)BacillomycinD,respectively.
