摘要
为测定常见食品中乳酸、富马酸的含量,建立了常见食品中乳酸、富马酸提取及反相高效液相色谱的测定方法。本实验选用葡萄酒、含乳饮料、糕点为基质,添加乳酸、富马酸标准品混匀,用水浸泡超声提取、稀释至刻度,摇匀,5000 r/min离心10 min,上清液过0.45μm无机滤膜后,以高效液相色谱反相C_(18)柱分离,通过PDA二极管阵列检测器检测。检测结果显示,以上3种基质用该测定方法检测的平均回收率分别为乳酸97.7%、95.5%、99.1%,富马酸96.3%、95.5%、97.9%;相对标准偏差分别为乳酸1.1%、1.5%、4.4%,富马酸1.2%、1.0%、0.3%。
In order to determine the content of lactic acid and fumaric acid in common foods,methods for the determination of lactic acid and fumaric acid extraction and reversed-phase high performance liquid chromatography in common foods were established.In this experiment,wine,milk-containing beverages,and pastries were used as the substrate,and lactic acid and fumaric acid standards were added and mixed,soaked in water and ultrasonically extracted,diluted to the mark,shaken,centrifuged at 5000 r/min for 10 minutes,and the supernatant was passed through a 0.45μm inorganic filter membrane.It was then used as a sample solution,and then separated by a reversed-phase C_(18) column of high performance liquid chromatography,and detected by a PDA diode array detector.The test results show that the average recoveries of the above 3 substrates detected by this method are lactic acid 97.7%,95.5%,99.1%,fumaric acid 96.3%,95.5%,97.9%;the relative standard deviations are lactic acid 1.1%,1.5%,4.4%,fumaric acid 1.2%,1.0%,0.3%.
