预针刺小鼠足三里对穴区皮肤及穴旁淋巴结记忆T细胞的影响

Effect of Preventive Acupuncture at Zusanli(ST-36) on Memory T Cells at Skin Tissue nearby Acupoint and Inguinal Lymph Nodes in Mouse

目的:观察预针刺小鼠足三里对穴区皮肤及穴旁腹股沟淋巴结记忆T细胞的影响,探讨预针刺在治未病方面的作用。方法:C57BL/6雄性小鼠按体质量随机分为空白组6只,预针刺组6只。预针刺组小鼠于足三里行针刺,30 min/次,1次/d,共10 d。治疗结束后用流式细胞术分别检测穴区组织驻留性记忆T细胞(TRM)、穴区CD3^(+)TRM、淋巴结CD3^(+)TRM、淋巴结CD3^(+)中央记忆T细胞(TCM)、CD3^(+)效应记忆T细胞(TEM)的细胞数量。结果:与空白组相比,预针刺可增加穴区组织CD69^(+)103^(+)TRM和CD3^(+)TRM数量(P<0.05),对穴区组织CD3^(+)TRM亚群和淋巴结CD3^(+)记忆T细胞(TM)亚群数量变化无明显影响(P>0.05);预针刺对穴区TRM/淋巴结TRM比值、淋巴结内TRM/TCM比值、淋巴结内TRM/TEM比值均无明显影响(P>0.05)。结论:预针刺可增加穴区组织CD69^(+)103^(+)TRM以及CD3^(+)TRM数量,这可能是针灸治未病的的物质基础之一。

Objective:To observe the effect of preventive acupuncture on Tissue resident memory T cells at Zusanli(ST-36)acupoint and inguinal lymph nodes in mouse,so as to explore the mechanism of preventive acupuncture in preventive treatment of Disease.Methods:Twelve C57 BL/6 male mice were randomly divided into blank group and preventive acupuncture group according to the weight.The mice in the preventive acupuncture group were given acupuncture at Zusanli(ST-36)for 30 minutes,once a day for ten days.After the preventive acupuncture treatment,the quantity of cells of TRM、CD3^(+)TRM at skin tissue at the acupoint area and CD3^(+)TCM、CD3^(+)TEM at inguinal lymph nodes were detected by flow cytometry.Results:Compared with the blank group,the quantity of cells of CD69+103+TRM and CD3^(+)TRM at skin tissue nearby the acupoint were significantly increased in the preventive acupuncture group(P<0.05).Statisticaly the quantity of cells of CD3^(+)memory T cell subset at inguinal lymph nodes and CD3^(+)TRM at the acupoint area were not significantly increased in the preventive acupuncture group(P>0.05).Preventive acupuncture has no effect on the ratio of TRM between acupoint skin tissue and inguinal lymph nodes(P>0.05)but can invert this radio.At the inguinal lymph nodes,preventive acupuncture has no effect on TRM/TCM and TRM/TEM(P>0.05).Conclusion:Preventive acupuncture can markedly increase the quantity of cells of CD69+103+TRM and CD3^(+)TRMat skin tissue nearby the acupoint,which may be one of the material bases for preventive treatment of Disease.
