中草药复方对断奶仔兔小肠发育及IL-2 mRNA表达量的影响

Effect of Chinese herbal medicine compound on small intestine development of weaned rabbits and IL-2 mRNA expression

摘要 为了探讨中草药复方对断奶仔兔肠道发育及肠道免疫水平的影响,试验选取30日龄、体重相近且健康状况良好的断奶伊拉兔仔兔60只,公母各半,随机分为2组,每组3个重复,每个重复10只,对照组饲喂基础日粮,试验组饲喂基础日粮+1%中草药复方,预试期5 d,试验期30 d。试验结束后,取小肠制作石蜡切片,观察小肠组织结构;测定空肠消化酶活性及小肠白细胞介素2(interleukin-2,IL-2)mRNA表达量。结果表明:(使用中草药复方后,试验组十二指肠、空肠和回肠的绒毛高度均较对照组显著增加(P<0.05),空肠的隐窝深度显著降低(P<0.05),三段肠段的绒毛高度与隐窝深度比值均极显著升高(P<0.01)。)试验组淀粉酶、胰蛋白酶活性较对照组显著或极显著提高(P<0.05或P<0.01);糜蛋白酶活性也有所提高,但差异不显著(P>0.05)。试验组空肠和回肠IL-2 mRNA表达量较对照组显著升高(P<0.05);十二指肠IL-2 mRNA表达量有升高趋势,但差异不显著(P>0.05)。说明中草药复方能够有效促进仔兔小肠发育,并增强其肠道免疫力。 In order to investigate the effects of Chinese herbal compound on intestinal development and intestinal immunity of weaned rabbits,60 Ira rabbits were weaned at the age of 30 days,with similar weights and in good health.They were divided into two groups randomly,each with three replicates,and each replicate with 10 rabbits.The control group was fed basal diet,and the experimental group was fed basal diet+1%Chinese herbal medicine compound.The pre-test period was five days and the test period was 30 days.After the experiment,small intestine slices were made to observe the morphology of the small intestine.Jejunal digestive enzyme activity and small intestinal interleukin-2(IL-2)mRNA expression level were measured.The results showed that after using the Chinese herbal medicine compound,compared with the control group,villus height of duodenum,jejunum and ileum in experimental group was significantly increased(P<0.05),crypt depth of jejunum was significantly decreased(P<0.05),villus height to crypt depth ratio of three intestinal segments were extremely significantly increased(P<0.01);Compared with the control group,the amylase and trypsin activities of the test group were significantly or extremely significantly increased(P<0.05 or P<0.01),and the chymotrypsin activity also increased,but the difference was not significant(P>0.05).The expression of IL-2 mRNA in the jejunum and ileum of the experimental group was significantly higher than that of the control group(P<0.05),and the expression level of IL-2 mRNA in the duodenum had a tendency to increase,but the difference was not significant(P>0.05).The results suggested that the Chinese herbal compound can effectively promote the development of the small intestine of the young rabbits and enhance their intestinal immunity.
