转化生长因子-β1在踝骨性关节炎软骨和血清中的表达

Expression of transforming growth factor-beta1 in serum and osteochondral tissue in ankle osteoarthritis

[目的]探索踝关节骨性关节炎(ankle osteoarthritis,AOA)血液和组织中转化生长因子β1(transforming growth factor-β1,TGF-β1)的表达。[方法]27例AOA患者行关节镜手术或踝关节融合,取血液和骨软骨组织检测TGF-β1。同期27例非A0A为血液检测对照,3例创伤截肢的骨软骨组织为免疫组化检测对照。[结果]ELISA法测定血清中TGF-β1.结果,AOA组(1994.70±188.49)pg/ml,对照组为(1623.22±127.99)pg/ml,两组间差异有统计学意义(P<0.001)。TGF-β1免疫组织化学染色A0A患者骨软骨标本中TGF-β1染色阳性,其中,15例染色较深,呈强阳性。软骨下骨中可见多核巨细胞,胞浆呈深蓝色。相比之下,正常软骨着色较淡,软骨下骨也可见多核巨细胞,但体积小,着色浅,[结论]AOA的血液和骨软骨组织TGF-β1表达显著高于正常对照。

[Objective]To explore the expression of transformed growth factor beta 1(TGF-β1)in blood and osteochondral tissues in ankle osteoarthritis(AOA).[Methods]The TGF-β1 was assessed in the blood and osteochondral tissue harvested from 27 patients who underwent arthroscopic surgery or ankle fusion for AOA.In addition,the blood samples taken from 27 non-AOA patients who were hospitalized for the other reason in the same period,and osteochondral samples from 3 cases of traumatic amputation without ankle involved were used as the normal controls.[Results]The TGF-β1 measured by ELISA in the serum was of(1994.70±188.49)pg/ml in the AOA group,whereas(1623.22±127.99)pg/ml in the control group,which was statistically significant(P<0.001).In term of inmniuohistochemistry stain-ing,TGF-β1 was positive in the AOA group,including the strong positive with darker staining in 15 cases.Polynuclear giant cells were found in the subchondral bone with dark blue plasma.In contrast,cartilage was lighter in the normal control group,with smaller and lighter polynuclear giant cell in the subchondral bone.[Conclusion]The AOA has considerably higher expression of TGF-β1 in blood and osteochondral tissue than the normal control.
