摘要
α/β水解酶结构域-6(ABHD6)是近些年来被发现的一种具有丝氨酸水解酶活性的分子,在中枢神经系统中发挥重要调控作用。作为突触后内源性大麻素的主要水解酶,ABHD6参与调节神经元内源性大麻素系统(ECS);此外,ABHD6还是α-氨基-3-羟基-5-甲基-4-异恶唑丙酸受体(AMPAR)复合体的重要组成部分,可抑制AMPAR向突触后膜转位和AMPAR的兴奋性。随着不断深入的研究,ABHD6在神经炎症通路和自身免疫调节中发挥着潜在的调控效应,有望成为干预神经系统疾病的新型靶点。该文从ABHD6的分子结构与表达、中枢神经系统中的作用机制等方面,对其在神经系统疾病治疗中的最新研究进展进行综述。
Recent studies have found thatα/β-hydrolase domain-containing 6(ABHD6)is a molecule that has the activity of serine hydrolase and plays an important regulatory role in the central nervous system.As the main hydrolase of postsynaptic endocannabinoid,ABHD6 is involved in the regulation of the neuronal endocannabinoid system;in addition,ABHD6 is an important part of theα-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor(AMPAR)complex and can inhibit the translocation of AMPAR to postsynaptic membrane and the excitability of AMPAR.In-depth studies show that ABHD6 plays a potential regulatory role in the neuroinflammation pathway and autoimmunity and is expected to become a new target for the intervention of nervous system diseases.This article reviews the latest research advances in the role of ABHD6 in the treatment of nervous system diseases from the aspects of its molecular structure,expression,and mechanism of action in the central nervous system.
