YB-1表达对宫颈癌细胞增殖的影响

摘要 目的探索YB-1的表达水平对宫颈癌细胞增殖的影响及其可能的机制。方法免疫组化检测宫颈鳞癌组织及癌旁正常组织中YB-1的表达水平,并分析其与临床病理间的关系;慢病毒转染将YB-1过表达载体转染至宫颈癌细胞SiHa中,RT-PCR检测转染效果。CCK-8实验和克隆形成实验检测YB-1过表达后SiHa细胞增殖和克隆能力变化情况;流式细胞术检测转染后SiHa细胞周期的变化。结果相对于癌旁组织,宫颈鳞癌组织中YB-1存在明显的过表达,且与组织学分型和淋巴结转移存在明显的相关性(P<0.05);CCK-8实验发现相对于对照组,YB-1过表达的SiHa细胞株的增殖和克隆能力明显增加(P<0.05);流式细胞术检测细胞周期发现YB-1过表达的SiHa细胞比例在G1期相对减少,S期相对增多,大约90%左右的细胞停滞在G1/S期。结论宫颈癌组织中YB-1存在高表达,在非角化型癌多见且更易出现淋巴结转移。YB-1的过表达能够促进宫颈癌SiHa细胞增殖。
