摘要
目的:探讨复方甘草酸苷对寻常型银屑病表皮细胞细胞周期的影响及其机制。方法:以不同浓度复方甘草酸苷(0.05、0.1、0.2 ng/ml)处理银屑病表皮细胞,采用流式细胞术分析细胞周期,Western blot测定细胞周期蛋白A(CyclinA)、细胞周期蛋白B1(CyclinB1)、细胞周期蛋白D1(CyclinD1)表达,qRT-PCR检测miR-155表达。采用脂质体将anti-miR-155转染至银屑病表皮细胞,qRT-PCR检测转染效果,流式细胞术、Western blot检测干扰miR-155对细胞周期的影响。结果:不同浓度复方甘草酸苷明显提高银屑病表皮细胞G0期细胞占比,显著降低S期细胞占比、CyclinA、CyclinB1、CyclinD1蛋白水平和miR-155表达,且呈浓度依赖性(P<0.05)。转染anti-miR-155成功干扰miR-155表达。干扰miR-155明显提高银屑病表皮细胞G0期细胞占比,显著降低S期细胞占比、CyclinA、CyclinB1和CyclinD1蛋白表达。miR-155可逆转复方甘草酸苷对银屑病表皮细胞周期的影响。结论:复方甘草酸苷可通过调控寻常型银屑病表皮细胞miR-155表达阻滞细胞周期。
Objective:To investigate effect of compound glycyrrhizin on cell cycle of epidermal cells of psoriasis vulgaris and its mechanism.Methods:Psoriatic epidermal cells were treated with different concentrations of compound glycyrrhizin(0.05,0.1,and 0.2 ng/ml),and cell cycle was analyzed by flow cytometry.CyclinA,CyclinB1 and CyclinD1 protein expressions were detected by Western blot.qRT-PCR to detect miR-155 expression.anti-miR-155 was transfected into psoriatic epidermal cells by liposomes,and qRT-PCR was used to detect transfection effect.Effect of interfering miR-155 on cell cycle was detected by flow cytometry and Western blot.Results:Compound glycyrrhizin at different concentrations significantly increased percentage of G0 phase cells of psoriatic epidermal cells,obviously decreased percentage of S phase cells,CyclinA,CyclinB1,CyclinD1 protein levels,and miR-155 expression in a concentration-dependent manner(P<0.05).Transfection of anti-miR-155 successfully interfered with miR-155 expression.Interfering with miR-155 greatly increased percentage of cells in G0 phase of psoriatic epidermal cells,and remarkably decreased percentage of cells in S phase,CyclinA,CyclinB1 and CyclinD1 protein expressions.miR-155 can reverse effect of compound glycyrrhizin on psoriatic epidermal cell cycle.Conclusion:Compound glycyrrhizin can block cell cycle by regulating expression of miR-155 in epidermal cells of psoriasis vulgaris.
